摘 要:目的 对腮腺液胆固醇检测方法进行探索和条件优化,探讨其与龋病发生的相关性.方法 采用Slominary方法收集不同龋敏感者的腮腺液,冷冻干燥后用Folch方法提取脂质.采用Alltech ODS作为预富集柱,采用反向HPLC 对腮腺液胆固醇进行定性、定量检测.结果 预富集流动相:乙腈/(甲醇/0.1 g/dl磷酸水溶液=6∶1)=5/95(V/V),流速0.3 ml/min;分析流动相:乙腈/甲醇=1/9 (V/V),流速1.0 ml/min,CS-HPLC优于HPLC,t=5.473,P〈0.05;高龋组腮腺液胆固醇含量明显高于无龋组.结论 CS-HPLC可以有效实现唾液痕量物质的预富集和净化,提高检测的灵敏度;腮腺液胆固醇与龋病具有相关性.Objective To explore the optimize of the methods of detecting of parotid cholesterol and to debate the relativity between cholesterol and caries. Methods The study was conducted with 10 caries-resistant subjects who had a DMFT (decayed missing and filled teeth) value of 0 and 8 caries-susceptible subjects who had a DMFT value of 〉5. Samples of parotid saliva (1 020 ml) were obtained and treated by the means of Slomiany B. L. Lipids were extracted and purified by the means of Folch. After dried under N2,the extractions were dissolved in methanol for further separation. The separation was done by an Allteeh ODS pre-column as pre-riehening column and Kromasil ODS column as analytical column. Results Solvent mixture of aeetonitrile/(methanol/0. 1 g/dl phosphoric acid aqueous solution=6/1)=5/95 (V/V) as the pre-richening mobile phase and acetonitrile/methanol= 1/9 (V/V) as analytical mobile phase respectively. The column efficiency of CSHPLC was higher than that of HPLC (t= 5. 473,P〈0.05). The amount of cholesterol of CR was considerably fewer than that of CS (P〈0.05). Conclusion The means of CS-HPLC for separation and purification of parotid salivary cholesterol significantly improved the sensitivity. The parotid cholesterol was correlated with caries.
