摘 要:目的 探讨酮替芬对大鼠急性前脑缺血再灌注损伤的保护作用及其机制。方法 采用Pulsinelli四动脉结扎法略加改良制作动物模型。记录EEG ,检测脑水分、脑组织内CK、MDA、SOD、Na+-K+-ATP酶和Ca2 +-ATP酶。结果 酮替芬可以促进模型鼠EEG的恢复 ,减轻脑水肿 ,使脑组织CK释放和MDA含量减少 ,提高SOD、Na+-K+-ATP酶和Ca2 +-ATP酶的活性。结论 酮替芬对脑缺血再灌注损伤有保护作用 ,可能与其减轻脑水肿 ,抗脂质过氧化 ,维持钠钾泵和钙泵功能有关。Objective To investigate the protective effect of ketotifen on acute cerebral ischemia-reperfusion in rats and the mechanism of its action. Methods Pulsinelli four-vessel occlusion method was used to make the cerebral ischemia-reperfusion model of rats. The EEG was recorded.The cerebral water content(BWC),CK and MDA contents of brain tissue,and the activity of SOD,Na +-K +-ATP enzyme and Ca 2+ -ATP enzyme were tested. Results By using ketotifen in model rats the recovery of EEG was promoted,BWC and CK contents were reduced and the activity of SOD, Na +-K +-ATP enzyme and Ca 2+ -ATP enzyme were improved. Conclusion Ketotifen protects the brain tissue from ischemia-reperfusion injury by anti-lipid peroxide reaction, protects the activity of Na +-K +-ATP enzyme and Ca 2+ -ATP enzyme and reduces brain edema and calcium accumulation.
