矢车菊素-3-葡萄糖苷在小鼠体内的吸收与代谢

摘　　要：【目的】研究花色苷矢车菊素-3-葡萄糖苷(C3G)在小鼠体内的吸收与代谢情况。【方法】48只小鼠随机分为7个实验组和1个对照组,每组6只。实验组小鼠灌胃C3G后测定不同时间点血浆、肝脏、肾脏、膀胱、胃及小肠中C3G、总花色苷和原儿茶酸(PCA)含量,分析其代谢产物。【结果】实验组小鼠血浆、肝脏、肾脏和尿液中均有C3G存在,在血浆中检测到了C3G的甲基化产物,肝脏、肾脏和尿液中检测到了C3G的甲基化、葡萄糖醛酸化和硫酸化产物。另外,在小鼠血浆、肝脏、肾脏、尿液和小肠中发现了PCA,在小肠中发现了矢车菊素,在肝脏和肾脏中检测到了矢车菊素的甲基化、葡萄糖醛酸化产物和少量香草酸。【结论】花色苷除以原型吸收外,部分在肠道可被代谢成酚酸,酚酸种类可能与B环上取代基有关。[Objective] To investigate the absorption and metabolism of cyanidin-3-glucoside （C3G） in mice. [Methods] Forty- eight mice were randomly divided into seven experimental groups and one control group, six mice in each group. Mice in experimental groups were garaged with C3G. In different time, plasma and urine were collected; liver, kidney, stomach, and small intestine were excised. C3G, total anthocyanins （ACN）, protocatechuic acid （PCA）, and metabolites of C3G were determined in all samples. [Results] C3G existed in the plasma, liver, kidney, and urine in experimental group mice. Moreover, peonidin-3-glucoside and delphinidin-3-glucoside were also detected in plasma. C3G methylation, C3G glucuronide, and C3G sulphate were detected in the liver, kidney, and urine. In addition, we found that PCA existed in the plasma, liver, kidney, urine, and small intestine after orally gavaged with C3G. Meanwhile, cyanidin was detected in the small intestine; cyanidin methylation, cyanidin glucuronide and a small amount of vanillic acid were detected in the liver and kidney. [ Conclusion ] A few anthocyanins could be absorbed by prototype, the others were metabolized to phenolic acids in the gut. The tvoe of ohenotic acids may be related to the substituent ~rouo of B cycle.
