摘 要:Nrf-2-ARE信号转导通路是机体最重要的抗氧化应激系统之一,在肿瘤的发生发展中扮演着双重作用,一方面可以预防肿瘤的发生;另一方面近期的研究发现,Nrf-2在肿瘤发生发展的过程中也起了重要作用。Nrf-2及其下游基因在许多癌前病变及肿瘤组织中表达异常升高,促进了异常增殖的细胞向肿瘤方向分化,肿瘤细胞的生长分裂能力增强,同时Nrf-2的升高能使肿瘤细胞对化疗药物敏感性下降,是肿瘤细胞耐药机制中的重要一环。然而,胃癌是威胁人类健康的恶性肿瘤之一,它的发生发展是一个涉及到分子和细胞改建的多步骤过程。由于受检测方法及技术的限制,目前早期胃癌检出率低,多数胃癌患者在确诊时已达中晚期,因此化学治疗在胃癌患者的综合性治疗中具有重要作用,但是化学治疗在肿瘤中的应用却存在着亟须攻克的问题-多药耐药。已有研究表明,Nrf-2在不同期胃癌、癌前病变中存在不同水平的高表达,对胃癌的发生发展有一定的促进作用,Nrf-2在细胞核中的堆积是一些肿瘤产生耐药性的原因。本资料就Nrf-2在胃癌发展过程中的作用及机制作相关阐述。Nrf-2-ARE signaling pathway is one of the body's most important anti-oxidative stress systems, which plays a dual role in the tumor occurrence and development. On the one hand, it can prevent tumor occurrence, on the other hand, recent studies have found that Nrf-2 also plays an important role in the process of tumor development. The expression of Nrf-2 and its downstream genes elevated abnormally in many precancerous lesions and tumor tissues, encouraging abnormally proliferous cells to differentiate toward tumor, enhancing the growth and division ability of tumor cells, and reducing the drug susceptibility of tumor ceils, which was an important step in the drug resistance mechanism of tumor cells. However, gastric cancer is one of the malignant tumors that threatens human health, of which the development is a multi-step process that involves molecular and cellular alterations. Due to the limitation of detection methods and techniques, the detection rate of early gastric cancer is low now and most patients with gastric cancer have reached middle and advanced stage before definite diagnosis, and therefore chemotherapy plays an important role in the comprehensive treatment gastric cancer, but the application of chemotherapy in gastric cancer encounters an urgent problem of muhidrug resistance. Studies have shown that Nrf-2 has different levels of high expression in different stages of gastric cancer and precancerous lesions, which has certain promoting effect on the occurrence and development of gastric cancer. The accumulation of Nrf-2 in the nucleus is the cause of some tumors' drug resistance. Therefore, this paper elaborates the role and mechanism of Nrf-2 in the development of gastric cancer.
