摘  要:目的观察艾灸对脾虚大鼠血清D木糖含量、空肠组织ATP含量及ATP酶活性的影响,探讨艾灸对ATP生成与膜蛋白含量的影响,分析艾灸温补脾胃改善脾虚证的作用机理。方法 40只SD大鼠随机分为4组:A空白对照组,B脾虚模型组,C艾灸穴位组(脾虚+艾条温和灸组),D中药对照组(脾虚+四君子汤组),每组10只。200%大黄水浸剂灌胃造成脾虚大鼠模型,艾灸足三里、中脘、关元、脾俞、胃俞等穴,间苯三酚法测定血清D-木糖含量,比色法测定空肠组织三磷酸腺苷(ATP)含量及Na+-K+-ATP酶(Na+-K+-ATPase)和Ca2+-Mg2+-ATP酶(Ca2+-Mg2+-ATPase)活性。结果与A组比较,B组脾虚模型大鼠血清D-木糖含量、空肠组织ATP含量、Na+-K+-ATPase和Ca2+-Mg2+-ATPase活性均显著降低(P<0.05或P<0.01);与B组比较,经艾灸足三里等穴或四君子药液灌胃治疗的C、D两组血清D-木糖含量、空肠组织ATP含量、Na+-K+-ATPase和Ca2+-Mg2+-ATPase活性均显著增高(P<0.05或P<0.01)。D组略优于C组,两组比较差异无显著性。结论艾灸能显著增加脾虚大鼠血清D-木糖含量,增加脾虚大鼠小肠组织ATP含量、ATP酶活性,提示艾灸可促进脾虚模型大鼠小肠上皮细胞ATP的生成,增加细胞膜蛋白的含量,改善物质跨膜转运,从而改善小肠吸收功能,促进脾虚症状恢复。[ Objective ] To observe the effect of moxibustion on content of D-xylose in serum, content of ATP and activities of ATPase in jejunal tissues of spleen-deficiency rats, and explore the effect of moxibustion on pro- duction of ATP and content of membrane proteins, to analyze the mechanism of moxibustion reinforcing spleen- stomach. [ Methods ] Forty healthy Sprague Dawley rats were randomly assigned into four groups, namely group A (blank group), B (model group), C (moxibustion group) and D (herbs group). The animal model of spleen-deficiency was established by intragastric administration with Chinese Rhubarb infusion. The rats in group A and B, and D served as the restraint, model, and intragastric administration with sijunzi decoction, respectively, while those in group C received moxibustion at zusanli, zhongwan, guanyuan, pishu and weishu points. The content of D-xylose in serum was detected by phloroglucinol method. Colorimetry method was used to detected content of ATP and activi- ties of Na+-K+-ATPase and Ca2+-Mg2+-ATPase in jejunum tissues. [ Results ] Compared with group A, the content of D-xylose in serum, content of ATP, activities of Na+-K+-ATPase and Ca2+-Mg2+-ATP in jejunum tissues were de- creased significantly (P 〈0.05 or P 〈0.01). Compared with group B, the content of D-xylose in serum, content of ATP, activities of Na+-K+-ATPase and Ca2+-Mg2+-ATP in jejunum tissues were increased significantly (P 〈0.05 or P 〈0.01). There was no significant difference between group C and D. [ Conclusions] Moxibustion could increase the content of D-xylose in serum and ATP in jejunum tissues, as well as activities of Na+-K+-ATPase and Ca2+-Mg2+- ATP of spleen-deficiency, which suggested that moxibustion could enhance absorption functions of small intestine to relieve spleen-deficiency symptoms by promoting the production of ATP in intestinal epithelial cells, increasing the content of membrane proteins, and improving transmembrane transport.
