摘 要:目的探讨槲皮素对高转移性人乳腺癌细胞MDA-MB-435S侵袭能力的影响及其相关机制。方法人乳腺癌细胞MDA-MB-435S以12.5、25、50、100和200μM终浓度的槲皮素处理后,台盼蓝拒染法检测细胞生长与增殖的变化;Transwell小室法检测细胞侵袭能力的变化;RT-PCR法及Western blot法分别检测基质金属蛋白酶(MMP)-9 mRNA水平和蛋白水平的表达变化。结果经槲皮素处理后,人乳腺癌细胞MDA-MB-435S体外增殖及侵袭能力均明显下降,MMP-9 mRNA和蛋白表达亦明显下调,且均与药物剂量呈正相关。结论槲皮素在体外剂量依赖性的抑制人乳腺癌细胞MDA-MB-435S的侵袭力,其机制可能与下调该细胞MMP-9的mRNA和蛋白表达有关。[ Objective ] To investigate the effect of quercetin on the invasive capacity of human breast cancer cell line MDA-MB-435S and its potential mechanism. [Methods ] Human breast cancer cells MDA-MB-435S were treated with different concentrations of quercetin (12.5, 25, 50, 100 and 200 μM), the proliferative capacity of the cells was measured using trypan blue dye exclusion test, the invasive capacity of the cells was measured using tran- swell chamber invasive model, and the expression levels of mRNA and protein of MMP-9 of the cells were detected by RT-PCR and western blotting assay respectively. [Results ] Quereetin could inhibit the proliferation and inva- sion and decrease the expression levels of mRNA and protein of MMP-9 of MDA-MB-435S cells, and showed the dose dependent relationships. [ Conclusions ] Quercetin could inhabit the invasion of human breast cancer cells MDA-MB-435 by dose dependent, and the underlying mechanism may be associated with down-regulation of MMP-9.
