摘 要:本实验通过石蜡切片染色观察统计比较水牛与黄牛卵巢各级腔前卵泡数量,并对水牛卵泡的超微结构进行了观察。结果表明:水牛卵巢中原始卵泡为37 578.81个,极显著高于初级卵泡的11 537.94个和次级卵泡的1753.40个(P<0.01);水牛卵巢腔前卵泡的总平均数为50 870.14个,极显著低于黄牛卵巢的113 483.32(P<0.01);超微结构观察发现水牛腔前卵泡卵母细胞和颗粒细胞间形成桥粒和间隙连接,细胞核位于胞质中心,细胞质中分布有大量的圆形脂质滴,存在粗面内质网以及少量游离的核糖体和皮质颗粒。实验从组织学水平上初步探明水牛比黄牛繁殖力低的原因,并为水牛腔前卵泡的分离培养提供了理论参考。The objective of the present study was to describe the quantity of preantral follicle of buffalo ovary, as well as the uhrastructural characterization of the preantral follicle. The results showed that the number of primordial follicle was estimated at 37 578.81 structures which was significantly different to primary and secondary follicles at 11 537.94 and 1 753.40 structures, respectively (P〈0.01). And the total preantral follicles of buffalo ovaries 50 870.14 was significantly lower than that of the bovine ovaries 113 483.32 (P〈0.01). The results of ultrastructure observation showed that there were some desmosome junctions and gap junctions between granulose cells and oocytes. The nucleus was central located, and there were small lipid vacuoles distributed evenly in the cytoplasm. Few cortical granules were visible in the region where contain well-developed rough endoplasmic reticulum and free ribosomes. The investigation provided the reason that buffalo have low reproduction ability than that of bovine, and provided scientific basis for utilizing oocytes from buffalo ovary preantral follicles in the future.
