摘 要:目的 观察高三尖杉酯碱 (HHar)诱导急性白血病 (AL)原代细胞凋亡的形态、生化特征 ,并分析量效关系 ,以指导临床治疗。方法 经 HHar作用后的 AL 原代细胞由光镜、电镜观察形态特征 ,DNA凝胶电泳、二苯胺法测定生化特征 ,SSPS软件分析量效关系。结果 HHar作用后 AL原代细胞形态、生化特征均符合典型的细胞凋亡改变 ;其DNA片段化率随 HHar作用时间的延长 ,浓度的增高而增加 ,具有时间——效应及剂量——效应线性趋势。DNA片段化率与坏死率正相关。结论 HHar确可诱导 AL 原代细胞凋亡。其诱导细胞凋亡的能力因剂量。Objective To observe the characteristics of apoptosis of primary actue leukemia cells induced by homoharringtoninum(HHar).Methods The morphological changes were observed by microscope, SEM and TEM, the biochemical characteristics were observed by DNA agarose gel electrophoresis, internucleosomal DNA fragmentation ratios. The time dose effect relationship was analyzed by SSPS software. Results The morphological and biochemical characteristics of apoptosis of primary acute leukemia cells induced by HHar were in common with the typical changes. The DNA fragmentation ratios of the primary leukemia cells increased gradully with increasing time and increasing HHar dose showing a time effect and dose effect linear trend, and was positively correlated to the necrosis rates of the cells. Conclusion HHar can induce primary acute leukemia cells apoptosis. Its effect is various according to different concentration and different time.
