中药体外抗幽门螺杆菌试验方法研究

摘　　要：目的探讨简便适用的中药体外抗幽门螺杆菌(Hp)试验的定性、定量方法。方法制备Hp选择琼脂平板,将中药药液倍比稀释后,制备含不同药物浓度的Hp选择琼脂平板,产气袋法培养Hp。先用打孔法定性测定中药制剂对Hp的抑菌作用,再用固体培养基连续稀释法和滴注法定量测定中药制剂对Hp的MBC。结果用4×105CFU/ml浓度菌液,1︰16稀释的试验药打孔法可测出抗Hp的抑菌圈;固体培养基连续稀释法和滴注法测得试验药对Hp的MBC为1︰40。结论利用打孔法、固体培养基连续稀释法和滴注法可测定中药制剂体外抗Hp效果。Objective To examine simple qualitative and quantitative methods to test the resistance of Helicobacter pylori to a traditional Chinese medicine in vitro. Methods Hp selective agar plates containing different concentrations of medicine were prepared after serial dilution. Hp was cultured using the GasPak system. The disk diffusion test was used to determine the inhibitory effect of a traditional Chinese medicine on Hp, and the serial dilution test on solid medium and instillation test were used to determine the MBC of the traditional Chinese medicine on Hp. Results A disk diffusion test with a 1 ： 16 dilution of the drug and 4×10^5 CFU/ml of bacteria resulted in an inhibition zone. The MBC of the drug on Hp was 1 ： 40 according to the serial dilution test on solid medium and instillation test. Conclusion The disk diffusion test, serial dilution test on solid medium, and instillation test can be used to determine action against HP of traditional Chinese medicine preparations in vitro.
