摘 要:目的 :分析慢性期慢粒白血病患者血清铜、锌、铁、镁含量与 bcl- 2、c- m yc基因表达水平的变化及其相互关系 ,为慢粒白血病发生机制研究及临床诊治提供实验依据。方法 :采用原子吸收分光光度法测定血清铜、锌、铁、镁含量 ;采用反转录 -聚合酶链式反应 (RT- PCR)及其扩增片段光密度扫描定量分析外周血白细胞中 bcl- 2、c-myc及内对照β- actin m RNA水平。结果 :与正常对照组比较 ,慢性期慢粒白血病患者血清锌及铁含量显著降低 ,血清镁含量及铜 /锌比值显著升高 ,血清铜含量升高但其差异无显著性 ;患者外周血白细胞中 bcl- 2及 c- myc m RNA水平均显著高于正常对照。结论 :慢粒白血病细胞中 bcr/ abl嵌合基因或其表达产物可致促进细胞增殖和 /或抑制细胞凋亡的相关基因表达增强 ,同时抑制锌、铁吸收和促进铜、镁的积累 ;血清铁、镁含量和铜 /锌比值反映出细胞增殖 -凋亡相关基因的表达水平 ,可能成为临床诊治慢粒白血病的重要参数。Objective:To investigate alteration of serum copper,zinc,ferrum and magnesium concentration and bcl 2、c myc gene expression level and the relationships between them in chronic phase patients with chronic myelogenous leukemia to provide experimental evidences for studying on pathological mechanisms and clinical diagnosis and/or treatment of chronic myelogenous leukemia.Methods:The concentrations of serum copper,zinc,ferrum and magnesium were detected by atomic absorption spectrophotometry.The mRNA levels of bcl 2、c myc and internal control β actin gene in peripheral white blood cells were quantitatively determined by reverse transcriptase polymerase chain reaction amplification for their mRNA and optical density scanning for the amplified cDNA fragments.Results:Comparing with that of the control,the concentrations of serum zinc and ferrum were significantly declined,and serum magnesium and copper/zinc ratio were significantly increased.The concentration of serum copper was higher than that of the normal control but there was no significant difference between them.The mRNA levels of bcl 2 and c myc in peripheral white blood cells from the patients were significantly higher than that of the cells from normal control.Conclusion:Bcr abl chimeric oncogene or its expression product could induce expression of the genes relating to promoting cell proliferation and /or suppressing apoptotic cell death,and in the meantime,it could inhibit assimilation for zinc and ferrum,and enhance accumulation of copper and magnesium.The concentrations of serum ferrum and magnesium and copper/zinc ratio could reflect expression levels of the genes relating to cell proliferation and apoptosis,furthermore,they'll probably be the important parameters for clinical diagnosis and evaluating for clinical treatment effects of chronic myelogenous leukemia.
