摘  要:目的:初步探讨IL-18基因转染治疗舌鳞癌的应用价值及抑瘤机制。方法:我们将人IL-18基因克隆至pcDNA3.1(+)并转染CRL1623细胞,以未转染细胞和空载体转染的CRL1623(pCDNA3.1/CRL1623)作为对照;随后在空载体pCDNA3.1(+)和pIL18转染后24,48和72h时,测定caspase3/7的活性;在空载体pCDNA3.1(+)和pIL18转染48h时,对IL-18和IFN-γ进行实时定量检测;SPSS16.0统计分析,使用t检验,P<0.05有统计学意义。结果:通过检测24,48和72h的细胞增殖情况发现,与非转染细胞、转染了空载体pcDNA3.1(+)的细胞相比,IL-18转染细胞caspase3/7活性显著提高。而且,与未转染细胞相比,IFN-γ在IL-18转染后48h的表达水平显著提高。结论:IL-18在调控舌鳞状细胞癌中扮演着重要角色,并可能在该癌症的治疗中起着重要的作用。Objective: To discusse the preliminary value and antitumor activity of IL--18 gene transfection for treatment of human tongue cancer. Methods: The CRL1623 cells were transfected with a eukaryotic expression vector encoding IL-18 gene named as pCDNA3.1/Interleukin-18, with the wild CRL1623 and the CRL-1623 transfected with empty vector (pCDNA3. 1/CRL1623) as controls. Then, we determined the number of viable cells in culture at 24, 48 and 72 h after transfection with pcDNA3.1(+) or piLl8. In order to analysis of cell growth, assessment of caspase 3/7 activity and real-time reverse transcription-polymerase chain reaction were used to assess expression of the ILl8 and IFNG (interferon-γ) genes. Statistical analyses were carried out using the SPSS statistical package, version 16.0 for Windows. The Student's t-test was used to compare the differences between groups. A P-value d0.05 was considered to be statistically significant. Results: Introduction of the ILl8 gene inhibited cell proliferation at 24, 48 and 72 h after transfection compared with untransfected cells and cells transfected with blank pcDNA3.1 (q-) vector. The numbers of apoptotic cells and caspase 3/7 activity were significantly enhanced by ILl8 transfection. Levels of IFN-γ and IL-18 mRNA were elevated after 48 h in ILl8-ransfected cells compared with wild-type cells. Conclusion: These findings suggest that IL-18 plays a role in the regulation of tongue squamous cell carcinoma and may represent a potential therapeutic target in this cancer.
