摘  要:目的探讨人重组骨形成蛋白-4(recombinant human bone morphogenetic protein-4,rhBMP-4)与人重组胰岛素样生长因子-Ⅰ(recombinant human insulin-like growth factor-Ⅰ,rhIGF-Ⅰ)联合应用对大鼠成骨细胞生长增殖及分化能力的影响。方法取大鼠颅盖骨组织块,采用改良组织块混合酶消化法培养成骨细胞,将第四代成骨细胞与10 ng/mL rhBMP-4(rhBMP-4组)、10 ng/mL rhIGF-Ⅰ(rhIGF-Ⅰ组)、10 ng/mL rhBMP-4加10ng/mLrhIGF-Ⅰ(联合组)、无血清低糖培养基(对照组)共同培养3 d,用噻唑蓝法测定细胞的增殖情况,用碱性磷酸酶试剂盒检测细胞碱性磷酸酶的活性,用羟脯氨酸试剂盒检测成骨细胞分泌Ⅰ型胶原的量。结果第3天时,4组促进大鼠成骨细胞增殖能力测定的光密度值差异具有统计学意义(F=4.080,P=0.016),碱性磷酸酶活性差异具有统计学意义(F=4.070,P=0.016),成骨细胞分泌Ⅰ型胶原的差异具有统计学意义(F=3.204,P=0.038);与对照组相比,rhBMP-4组,rhIGF-Ⅰ组及联合组,都可增强成骨细胞的增殖分化能力,但rhBMP-4和rhIGF-Ⅰ联合应用不比单独应用的作用强。结论 rhBMP-4与rhIGF-Ⅰ联合应用,不能协同促进大鼠成骨细胞增殖及分化能力。Objective To study the effect of recombinant human bone morphogenetie protein (rhBMP-4) and recom- binant human insulin-like growth factor- I (rhlGF- I ) on the proliferation and differentiation of rat osteoblasts. Meth- ods Rat osteoblasts were treated with 10 ng/mL rhBMP-4, 10 ng/mL rhlGF- I , 10 ng/mL rhBMP-4 plus 10 ng/mL rhlGF- I for 3 days, the proliferation was mesured by MTF colorimetric essay, the ALP activity was mesured by ALP kit, type I collagen in osteoblasts were mesured by hydroxyprolline kit. Results There were significant difference between experimental groups of rhBMP-4, rhlGF- I , rhBMP-4 plus rhlGF- I and the control group in the proliferation and differ- entiation of cells in 3 days, but there were no differences among the three experiment groups. Conclusion There were no synergistieal effect of rhBMP-4 plus rhlGF- I on the proliferation and differentiation of rat osteoblasts.
