摘　　要：目的探讨缺血再灌注大鼠缺血脑组织fibulin-5的表达规律。方法 96只雄性SD大鼠分为正常组、假手术组、中脑动脉闭塞(MCAO)模型6、24、48和72 h组。线栓法制备局灶性脑缺血再灌注模型,激光共聚焦定位fibulin-5在脑组织内的表达,Western印迹检测缺血后fibu-lin-5蛋白表达变化,实时定量PCR测定fibulin-5 mRNA表达规律。结果①fibulin-5主要在内皮细胞间表达,偶可在实质细胞或内皮细胞胞质内表达;②大鼠缺血侧脑组织fibulin-5蛋白表达从再灌注6 h开始升高(1.26±0.46,P<0.05),再灌注24 h达到高峰(1.78±0.48),72 h(0.89±0.27)时有所降低,但仍高于假手术组(0.30±0.14,P<0.05),fibulin-5 mRNA表达规律与蛋白表达有同样的趋势。结论缺血再灌注大鼠缺血脑组织内fibulin-5表达水平明显升高,有可能对脑损伤后血管稳定起到保护作用。Objective To observe the expression of fibulin-5 in rats after cerebral ischemia reperfusion（I/R）.Methods 96 male Sprague-Dawley rats were randomly divided into 6 groups：control,sham-operation,and 6,24,48,72 h after I/R groups.The I/R model was induced by ligation with nylon monofilament.The expression of fibulin-5 was assessed by real-time quantitative PCR,Western blot and Laser confocal.Results Results ①The protein expression of fibulin-5 was significantly increased at 6 h after I/R（1.26±0.46,P0.05）,and it reached the summit at 24 h（1.78±0.48） after reperfusion,it was lower than that in 24 h group at 72 h（0.89±0.27） after injury,but still higher than that in sham-operation group（0.30±0.14,P0.05）.The expression of fibulin-5 mRNA had the same trend.②The fibulin-5 was mainly expressed between endothelial cells.Conclusions The expression of fibulin-5 is significantly increased in brain of rats after I/R,and this possibly play a role in protection of vascular stability after brain injury.
