丙戊酸钠和拉莫三嗪对雌性癫痫大鼠血清性激素水平的影响

摘　　要：目的探讨丙戊酸钠(Valproate sodium,VPA)和拉莫三嗪(lamotrigine,LTG)对雌性癫痫大鼠性激素水平的影响。方法 28只8周龄SD雌性大鼠随机分为四组:正常对照组、模型组、丙戊酸钠组(VPA组)和拉莫三嗪组(LTG组)。模型组、VPA组、LTG组用海人酸(KA)点燃,建立癫痫大鼠模型。建模成功后灌胃法分别予以生理盐水、丙戊酸钠、拉莫三嗪1个月,每3d称重1次,调整给药剂量。灌胃30d后,于动情间期四组大鼠抽取动脉血,电化学发光法测定血清黄体生成素(Luteotropic hormone,LH)、卵泡刺激素(Follie stimulating hormone,FSH)、睾酮(Testosterone,T)、孕酮(Progesterone,P)、雌二醇(Estradiol,E2)浓度,同时取双侧卵巢组织,卵巢称重,行HE染色,运用图像分析软件Image-pro Plus 6.0进行形态学分析。结果模型组与对照组比较,血清睾酮、黄体生成素水平升高(P<0.05),孕酮水平降低(P>0.05);模型组和VPA组大鼠失去正常动情周期;VPA组大鼠体重增加量较其它三组显著增高(P<0.01);VPA组与对照组比较,血清黄体生成素和睾酮升高(P<0.05),卵泡刺激素和孕酮降低(P<0.05);LTG组大鼠激素水平较对照组差异无统计学意义(P>0.05);VPA组与LTG组大鼠激素水平比较,差异有统计学意义(P<0.05)。结论雌性癫痫模型大鼠性激素水平异常,丙戊酸钠较拉莫三嗪易引起雌性癫痫大鼠性激素分泌紊乱。Objective To detect the serum concentrations of folliele stimulating hormone （ FSH）, Luteotropic hormone （LH） , estradiol （ E2 ） , Progesterone （P） and Testosterone （T） of epileptic female rats induced byKainie acid （KA） and to explore the effects of valproate and lamotrigine acid on serum sex hormone levels in female rats with epilepsy. Methods 8 - week - old Sprague - Daweley 28 rats were divided randomly intofour groups. Controls were fed normal saline 2mL every day and rest groups were established epileptic modle induced by Kainic acid（ KA）, randomly administered with VPA, LTG and NS by intragastric administration for30 days. Weight was monitored per 3 days and dose was adjusted according to body weight. Estrous cycles were monitored with daily vaginal smears. After 30 days, Bleeding, separating of ovary organs in their diestrus. SerumFSH, LH,E2, P and T level were determined using the electrochemical luminescence method . Results VPA group showed significant weight gain （P 〈0.01 ）. The level of LH was significantly increased in the VPA andNS groups compared with control group. The level of FSH was significantly decreased in the VPA group compared with the control and LTG groups, the level of T was significantly increased in the VPA group comparedwith LTG and control groups . The level of P was significantly decreased in the VPA group compared with control, LTG and NS groups. Conclusion Valproic acid sodium is easier to cause sex hormone secretion disorderthan lamotrigine.
