摘 要:目的观察p53、bcl-2、bax在甘氨脱氧胆酸盐诱导人正常肝细胞HL-7702细胞凋亡中的表达,并探讨其凋亡机制。方法终浓度分别为100、150、200、250μmol/L的GCDC处理HL-7702细胞24h,AnnexinV-FITC/PI双染色法检测GCDC诱导HL-7702细胞凋亡的凋亡率,RT-PCR测定p53、bcl-2、bax基因mRNA表达水平。结果 100、150、200、250μmol/L的GCDC均可诱导HL-7702细胞凋亡,呈剂量依赖性,凋亡率分别为(13.16±2.9)%(t=6.41),(20.3±3.0)%(t=10.22),(25.02±2.1)%(t=18.11),(45.02±3.5)%(t=28.89),较对照组(2.2±0.6)%显著升高(P<0.05);GCDC使细胞内bcl-2 mRNA的表达下降、p53和bax mRNA的表达增加。结论 p53、Bcl-2、Bax在甘氨脱氧胆酸盐诱导人正常肝细胞HL-7702细胞凋亡中发挥重要作用。Objective To study on the effect of p53、bcl-2、bax in apoptosis of cultured human hepatocyte HL-7702 induced by glycodeoxycholate(GCDC) and to explore its possible mechanism.Methods HL-7702 cells were incubated with various concentrations(100、150、200、250μmol/L) of GCDC for 24h.The apoptotic rate of HL-7702 was determined by AnnexinⅤ-FITC/PI double staining.The mRNA expression levels of p53、bcl-2/bax in HL-7702 cells were analyzed by RT-PCR.ResultsHL-7702 cells could be induced to undergo apoptosis in a concentration dependent manner after 100μmol/L、150μmol/L、200μmol/L、250μmol/L GCDC treatment for 24 hours.The apoptotic rates were(13.16±2.9)%,(20.3±3.0)%,(25.02±2.1)%and(45.02±3.5)%,which were markedly higher(P〈0.05)than that of the control group.The expression of p53、bax mRNA were increased and the expression of bcl-2 mRNA was decreased.Conclusion p53、bcl-2 and bax play an important role in apoptosis of cultured human hepatocyte HL-7702 induced by GCDC.
