摘 要:【目的】为研究microRNA在猪肌肉生长发育中的作用。【方法】采用Illumina高通量测序技术对猪背最长肌和腰大肌的microRNA转录组进行测定,并鉴定差异表达的microRNA。【结果】在猪背最长肌和腰大肌文库中分别产生了15.22M和17.52M的序列;两个文库共检测到的695个microRNA,其中有363个共表达、193个差异极显著(P<0.01)。【结论】说明microRNA在不同骨骼肌类型中存在差异表达。【Object】This experiment was conducted to investigate the effects of MicroRNAs in the development of skeletal muscle.【Method】Illumina Solexa sequencing technolog were applied to acquire the miRNA expression patterns between porcine longissimus doris muscle and psoas major muscle. 【Result】a total of 15.22M and 17.52M mappable sequences were expressed in longissimus doris muscle and psoas major muscle library respectively.Totally 695 unique miRNAs were observed, and 363 were co-expressed in two libraries, of which 193 were significantly differentially expressed between longissimus doris muscleand and psoas major muscle library (P〈 0.01). 【Conclusion】All these differences indicated the essential roles of miRNAs in the phenotypic variations of different muscle fiber types.
