摘 要:【目的】优化醇碱法提取厚朴总酚(和厚朴酚、厚朴酚)的工艺条件。【方法】以和厚朴酚、厚朴酚、厚朴总酚得率为指标,考察乙醇浓度、碱浓度、料液比、超声时间对提取效率的影响,采用响应面法分析各因素的影响程度和各因素间的相互作用。【结果】建立的二次回归方程与试验数据拟合度高、预测性好,和厚朴酚、厚朴酚、厚朴总酚的R2分别为0.940 2、0.962 6、0.960 6。经响应面分析,在超声强度150W下,提取厚朴的最佳条件为乙醇浓度56%、碱浓度0.8%、料液比1∶36、超声时间29min,在此条件下和厚朴酚、厚朴酚、厚朴总酚得率分别为1.95、9.80、11.75mg/g。【结论】所建立的回归方程对厚朴提取条件的优化是可行的,优选出的最佳工艺具有提取得率高、时间短、耗能少等优点。【Objective】 The optimum extraction conditions for highest yields of total phenols (magnolol, honokiol) were analysed for cortex Magnolia officinalis var. biloba, using the response surface analysis. 【Methods】 The extraction factors of the concentration of ethanol, concentration of alkali, solid-to-liquid and periods of ultrasound-assisted extraction were instigated. 【Results】 The second-order quadratic regression equation fitted the experimental data well with regard to magnolol (R2=0.9402), honokiol (R2=0.9626) and total phenols (R2=0.9606), respectively. Under the condition of ultrasonic power 150 W, the optimum extraction conditions for total phenols were ethanol 56%, alkali 0.8%, solid-to-liquid 1∶36, period of extraction 29 min, where herein yields of magnolol were 1.95 mg?g-1, honokiol were 9.80 mg?g-1, total phenols were 11.75 mg?g-1. 【Conclusion】 The predicted values got consistent with experimental values. Ethanol-alkali extraction of total phenols dominated in time, power and efficiency, comparing with ethanol and alkali extraction.
