Flt3配体联合IL-15诱导小鼠脾脏NKDC功能的实验研究

摘  要:目的探讨Flt3配体联合IL-15提高小鼠脾脏自然杀伤性树突细胞(natural killer dendritic cell,NKDC)杀伤肿瘤细胞的活性和增强刺激小鼠T细胞增殖的功能。方法将C57BL/6小鼠分为4组:A组不予任何因子处理,B组腹腔注射小鼠重组F1t3配体(100 ng/只),C组腹腔注射小鼠IL-15(500 ng/只),D组同时腹腔注射F1t3配体(100 ng/只)和IL-15(500 ng/只)。分别检测不同处理组小鼠脾脏NKDC杀伤小鼠淋巴瘤Yac-1细胞的活性、刺激小鼠T细胞增殖的功能及培养上清液中IFN-γ的分泌。结果在单独F1t3配体或单独IL-15作用下,NKDC杀伤小鼠淋巴瘤Yac-1细胞的活性与对照组比较差异无统计学意义(65.7%、66.5%vs 65.0%,P=0.12、P=0.13),与对照组比较,F1t3配体联合IL-15显著提高NKDC杀伤小鼠淋巴瘤Yac-1细胞的活性(78.8%,P=0.03);F1t3配体作用下NKDC刺激T细胞活性增强(为对照组的1.6倍),而在IL-15作用下,刺激T细胞活性降低(仅为对照组的0.8倍)。在F1t3配体和IL-15联合作用下,其刺激T细胞活性较对照组高(为对照组的1.5倍),略低于F1t3配体的作用。F1t3配体单独作用下,细胞上清IFN-γ为(97.0±0.5)μg/L,与对照组(98.5±1.2)μg/L比较差异无统计学意义(P>0.05),而在IL-15作用下或在F1t3配体和IL-15联合作用下,其IFN-γ分泌均有显著提高,在IL-15作用下,其IFN-γ分泌为(980±3.2)μg/L,而在F1t3配体和IL-15联合作用下,IFN-γ分泌为(991±2.5)μg/L,与对照组比较,差异均有统计学意义(P=0.008,P=0.007)。结论 F1t3配体联合IL-15可增强NKDC细胞的杀伤活性和刺激T细胞增殖的功能及增加NKDC培养上清中IFN-γ的分泌。Objective To investigate the effect of Fh3 ligand and IL-15 on the function of murine spleen natural killer dendritic cells (NKDCs). Methods C57BL/6 mice were divided into 4 groups:mice in Group A were injected with normal saline, Group B with Flt3 ligand ( 100 ng per mouse) ,Group C with murine IL-15 (500 ng per mouse) and Group D with Flt3 ligand( 100 ng per mouse) + IL-15 (500 ng per mouse). The spleen NKDCs were isolated and cultured in vitro. The eytolytic activity of NKDCs against Yac-1 cells and the capacity of T cell stimulation were assayed; the levels of IFN-γ secreted by NKDCs were detected. Results The cytolytic activity of NKDCs against Yac-1 cells in Group D ( Flt3 + IL-15) was higher than that in Group A (control) (78.8% vs 65.0% ,P 〈 0. 05 ) ,however,that in Group B ( Flt3 ligand) and C ( IL-15 ) was 65.7% and 66.5% ,which was not significantly different with that in Group A. The stimulatory activity for murine spleen T cells was increased in Group B and decreased in Group C compared to Group A. The stimulatory activity in Group D was higher than that in Group A, but little lower than Group B. Compared to Group A, the IFN-γ, production in Group C was increased from (98.5 ± 1.2)μg/L to (980 ±3.2) μg/L. Similar increase was observed in Group D. There was no signifieant difference in IFN-γproduction between Groups B and A. Conclusion Combined treatment of Fh3 ligand and IL-15 can enhance the cytolytie activity and T cell stimulation ability of murine spleen NKDCs.
