摘  要:目的探讨罗布麻提取物(AVE)对大鼠局灶性脑缺血再灌注损伤的防护机制。方法 50只Wistar雄性大鼠随机分为假手术组、模型组、罗布麻提取物中剂量(25 mg.kg-1.d-1)组、罗布麻提取物高剂量(50 mg.kg-1.d-1)组、银杏叶胶囊对照组。给药组术前连续灌胃5 d,于末次给药30 min后行手术,采用线栓法制备大鼠大脑中动脉缺血模型,缺血2 h再灌注24 h后进行神经病学评分;氯化三苯四氮唑(TTC)染色检测脑组织梗死面积;免疫组织化学检测Bcl-2和Bax蛋白表达。结果药物干预组与模型组比较评分及脑组织梗死面积均差异显著(P<0.05);药物干预组Bcl-2蛋白的表达上调,Bax蛋白的表达下调。结论与模型组相比,2个AVE剂量组可以减轻神经症状,缩小缺血面积;AVE可增强Bcl-2蛋白表达,下调Bax蛋白表达,从而抑制脑缺血再灌注后神经细胞凋亡,这可能是AVE对脑缺血再灌注损伤的神经保护作用机制之一。Objective To investigate the effects of apocynum venetum L extract(AVE)on neuron apoptosis after cerebral ischemia and reperfusion in rats.Methods 50 Wistar male rats were randomly divided into sham operation group,untreated group,medium dose AVE treated group(25 mg·kg-1·d-1),high dose AVE treated group(50 mg·kg-1·d-1),and ginkgo leaf capsules treated group.The animals of medicine groups were intragastrically administered with drugs for 5 days.30 minutes after the end administration,the operation was performed to induce the middle cerebral artery occlusion(MCAO)model in rats with the method of intraluminal thread.The middle cerebral artery blood flow was blocked for 2 hours and reperfused for 24 hours,then the rats'neurology score was observed.Brain triphenyltetrazolium chloride(TTC)staining and Immunohistochemical method were conducted to observe the effect of Bcl-2 and Bax expression.Results The indexes mentioned above in the infarct sizes of rat brain tissue in the AVE treated groups obviously decreased compared with those of the untreated group(P0.05).Drug groups could markedly upregulate the expression of anti-apoptosis gene Bcl-2 and restrain the expression of Bax.Conclusion AVE could inhibit neuron apoptosis by improving the expression of Bcl-2 protein and down-regulating the expression of Bax protein,which maybe one of the mechanisms of neuron protective effects of AVE on cerebral ischemia and reperfusion injury.
