摘  要:通过采用水提醇沉、三氯乙酸脱蛋白、分级醇沉等方法对松茸多糖提取物进行初步分离纯化,获得9个组分。用凝胶渗透色谱-十八角度激光散射联用系统和酪氨酸酶抑制法对9个组分的分子质量、多糖纯度和美白活性进行测定。通过纯度和美白功效选择最优的组分通过凝胶层析法分离纯化。结果表明,松茸多糖中分离得到TMSP-6Ⅱ,重均分子质量Mw=7.251×103D,纯度不小于95%,其具有良好的抑制酪氨酸酶活性,IC50值为136.4 mg·L-1。Tricholoma matsutake pelysaccharide extract was obtained from Tricholoma matsutake Sing with methods including water extraction and alcohol precipitation. Then the extract was preliminary separated and purified via protein removal with trichloroacetic acid treatment and fractional alcohol precipitation, and nine ingredients were obtained. Molecular weight, polysaccharide purity and whitening efficacy of the nine ingredients were determined and identified by gel permeation chromatography - multi - angle laser light scattering detector system and tyrosinase inhibition test. According to test result for purity and whitening efficacy,the best ingredient was chosen and further separated and purified with gel chromatography. Result indicated that the ingredient TMSP -6 II isolated from Tricholoma matsutake polysaccharide displays good tyrosinase inhibition activity. Its weight - average molecular weight was determined as 7. 251 ×10^3 D and its purity was determined as higher than 95% with IC50, value determined as 136.4 mg · L-1.
