摘 要:目的观察粉防己碱(tetrandrine,Tet)联合阿霉素(adriamycin,ADM)对人膀胱癌BIU-87/ADM细胞裸鼠移植瘤抑制率的影响。方法 30只裸鼠腋部皮下接种BIU-87/ADM移植瘤细胞,第22天扪及一小结节,随机分为对照组(生理盐水;6只)、ADM组(1.0mg/kgADM;6只)、Tet低剂量联合ADM组(1.0μg/mlTet+1.0mg/kgADM;6只)、Tet中剂量联合ADM组(3.0μg/mlTet+1.0mg/kgADM;6只)和Tet高剂量联合ADM组(5.0μg/mlTet+1.0mg/kgADM;6只),每4天于肿瘤局部注射生理盐水、ADM以及不同剂量的Tet+1.0mg/kgADM,每4天测量肿瘤大小,并绘制肿瘤生长曲线;药物处理结束后取瘤并称取重量,依据公式计算抑瘤率。结果 Tet各剂量联合ADM组肿瘤的体积、瘤重与对照组比较,差异有统计学意义(P<0.05);Tet中、高剂量联合ADM组肿瘤体积、瘤重与ADM组比较,差异有统计学意义(P<0.05)。结论 Tet联合ADM能够明显增加ADM对膀胱癌细胞BIU-87/ADM移植瘤杀伤的敏感性,Tet剂量越高,抑制作用越明显。Objective To investigate the effect of drug-combination of tetrandrine (Tet) and adria- myein (ADM) on the inhibition rate of BIU-87/ADM muhidrug resistance tumor xenografts. Methods 30 nude mice were inoculated axillary subcutaneously BIU 87/ADM tumor cells, a palpable nodules was founded after twenty two days, randomly divided into 5 groups: control group (NS group, n= 6), ADM group (1.0 mg/kg ADM, n 6), low-dosage Tet and ADM group (1.0μg/mi Tet +1.0 mg/kg ADM, n 6), middle-dosage Tet and ADM group (3.0μg/ml Tet+l.0 mg/kg ADM, n--6), high-dosage Tet and ADM group (5.0μg/ml Tet +1.0 mg/kg ADM, n=6). Every four days in the local tumor injection of NS, ADM and different doses of Tet + 1.0 mg/kg ADM. Measuring tumor size every four days, and mapping tumor growth curve; after drug treatment, taking the tumor and weighting; and calculaing the in- hibitory rate according to formula. Results In the groups of the combination of the ADM and Tet with dosage classified by three types (high, middle, low), the volume and weight all showed statis- tical significance compared with the NS control group (P〈0.05). In the groups of high-dosage and middle-dosage, the inhibition of tumor exnografts also showed statistical significance compared with ADM positive control group (P〈0.05). Conclusions The drug-combination of Tet and ADM can enhance the sensitivity of muhidrug resistance cells BIU-87/ADM to the doxombicin, the dose of the tetrandrine higher, the obvious inhibition more.
