水稻外源DNA导入变异系与受体的农艺性状差异及其基因变异分析

摘　　要：野威B是将小粒野生稻基因组DNA通过穗茎注射法导入杂交水稻保持系V20B后创造的变异系,其在株型、稻米品质和产量相关性状上都发生了明显变化。通过全基因组测序,比较分析了野威B与V20B中27个可能与变异性状有关的基因,发现野威B在与株型相关的基因中存在21个SNP和30个InDel,其中有10个SNP和4个InDel位于编码序列;在稻米品质相关的基因中存在124个SNP和28个InDel,其中25个SNP位于编码序列;在产量相关基因中存在65个SNP和42个InDel,其中有10个SNP和2个InDel位于编码序列。位于编码序列的变异中,有24个SNP为错义突变,导致部分编码氨基酸的极性或电荷状态改变,有4个InDel变异为移码突变,导致编码的氨基酸序列改变。为寻找引起变异系野威B表型变化的关键基因及位点提供依据。Yewei B （YVB） is a variant line from V20 B, a popular maintainer line in hybrid rice, by introducing genomic DNA of the wild rice of O. minuta through the spike-stalk injection method. Compared to its receptor V20 B, YVB has apparent changes in agronomic characters including the traits of plant type, grain quality and grain yield. Based on the whole genome sequencing, 27 genes related likely to the varied traits were compared and ana- lyzed between YVB and V20B. The results showed that there were 21 SNPs and 30 InDels in YVB in the genes of plant type traits, of which 10 SNPs and 4 InDels fell into the coding sequence （CDS）. In the genes of grain quality traits, there were 124 SNPs and 28 InDels in YVB, of which 25 SNPs were in CDS, and in the genes of grain yield traits, there were 65 SNPs and 42 InDels in YVB, of which 10 SNPs and 2 InDels occurred in CDS. Among all variations in CDS, 24 SNPs belonged to missense mutations, resulting in changes in the polarity or charge status of some amino acids, and 4 InDels belonged to frameshift mutations, resulting in changes in amino acid sequence. This study provides the basis for locating key genes of phenotypic changes in the variant line YVB.
