摘 要:目的探讨BMSCs移植对老年痴呆大鼠空间记忆能力的改善情况,为细胞移植治疗老年痴呆实验依据。方法海马注射Aβ1-40蛋白制备AD动物模型,模型成功后移植BMSCs。Morris水迷宫检测大鼠行为学变化,采用BrdU、NSE和GFAP的免疫荧光双标技术观察移植的细胞生存及分化。结果 AD组大鼠逃避潜伏期较对照组明显延长(P<0.05),移植BMSCs治疗后大鼠逃避潜伏期明显缩短(P<0.01);移植细胞后大鼠海马周围可见分化为NSE或GFAP阳性的细胞。结论 BMSCs移植AD大鼠后能在局部存活并分化,并且能够改善AD大鼠空间学习记忆能力。Objective To study the improvement of spatial memory ability after transplantation of bone marrow mesenchymal stem cells (BMSCs) in rats with Alzheimer's disease, and provide experimental data for cell transplantation treatment. Methods AD model was induced by injecting Aβ1-40 protein into hippocampus. When the model succeeded, the BMSCs were implanted into the hippocampus area. The behavioral changes of rats were evaluated with Morris water maze. BrdU, NSE and GFAP immunofluorescence double-staining was adopted to test the BMSCs survival and differentiation. Results The escape latency of hidden platform in AD group prolonged significantly (P 〈0.05), the escape latency after transplantation BMSCs shortened obviously compared with AD group (P 〈0.01). The BrdU/NSE and BrdU/GFAP double staining positive cells could be found surround hippocampus in rats of transplantation group. Conclusions The BMSCs can survive and differentiate after transplantation into the rats, and maybe improve the spatial learning and memory ability of AD rats.
