陈旧性心肌梗死大鼠的实验研究

摘　　要：[目的]观察陈旧性心肌梗死大鼠的病理变化及细胞凋亡情况。[方法]40只SD雄性大鼠被随机分为模型组和对照组,每组20只,模型组结扎冠状动脉左前降支造成急性心肌梗死模型,对照组开胸不结扎,术后3个月留取心脏标本,2,3,5-氧化三苯基四氮唑(tripheng ltetrazoliurn chlo-ride,TTC)染色法检测心肌梗死面积,HE染色法观察病理改变,Western blot法检测Caspase-3蛋白表达量。[结果]对照组梗死面积为0,Caspase-3蛋白表达量为(0.31±0.09),模型组的梗死面积为(30.60±5.20)%、Caspase-3蛋白表达量为(0.49±0.08),模型组显著高于对照组,P<0.01;HE染色发现模型组心肌细胞稀疏、萎缩,间质组织纤维化伴慢性炎症细胞浸润,心内膜软骨化,形态学特征符合透明软骨细胞。[结论]陈旧性心肌梗死大鼠梗死边缘区存在细胞凋亡。[Objective] To investigate the histopathological change and cell apoptosis of rats with chronic myocardial infarction.[Methods] 40 male Sprague-Dawley（SD） rats（weight 180~220g） were randomly divided into 2 groups： model group and sham group.Model groups underwent coronary ligation of left anterior descending branch to establish acute myocardial infarction models,and sham group only received thoracotomy without coronary ligation.3 months after operation,myocardium tissue was collected to measure the infarction size by TTC staining,detected histopathological change by HE staining and protein expression levels of Caspase-3 by western blot.[Results] Comparing with rats in sham group,the infarction size（0 vs 30.60±5.20%） and Caspase-3 expression（0.31±0.09 vs 0.49±0.08） were higher in rats in model group.HE staining in rats of model group shown in the myocardium region cardiocytes shrank and dispersed.Increasing fibrosis,edema of interstitium tissue,infiltration of few chronic inflame cells and many dilated congestive capillaries were visible.In the endocardium region,endocardium became thick obviously.Cardiocytes vanished and interstitium tissue went fibrous.Mature and immature chondrocytes gathered and formed many nests of chondrocytes.Judging by morphology,they belonged to hyaline cartilages.[Conclusion] In infarcting border zone,myocardial cell apoptosis existed in rats with chronic myocardial infarction.
