摘  要:目的建立酸中毒诱导的早产儿视网膜病(ROP)大鼠动物模型(AIR模型),研究血管内皮生长因子(VEGF)、血管生成素-2(Ang-2)的表达变化规律,探讨其与ROP血管发育及新生血管形成之间的关系。方法 1日龄SD新生大鼠随机分为酸中毒诱导模型组(ROP组)和正常对照组(C组),通过管饲NH4Cl方法建立AIR模型。应用免疫组化方法标记视网膜VEGF、Ang-2的表达水平。结果免疫组化结果显示,8日龄ROP大鼠VEGF、Ang-2的表达水平明显低于对照组(P<0.05),而在停止管饲NH4Cl后的10日龄、13日龄ROP组大鼠视网膜VEGF、Ang-2表达水平明显高于对照组(P<0.05)。在实验过程中VEGF及Ang-2的表达趋势相同。结论酸中毒使VEGF及Ang-2的蛋白表达水平下调;停止管饲NH4Cl后2者表达水平上调;2者变化过程与视网膜新生血管发展进程有关。VEGF、Ang-2可能在视网膜正常血管发育及ROP的新生血管化进程中起到重要作用。Objective To establish an animal model of acidosis-induced retinopathy ( AIR), analyze the expression of vascular endothelial growth factor (VEGF) and angiopoietin-2 (Ang-2) in a rat model of acidosis-induced retinopathy and to evaluate the relationship between VEGF, Ang-2 and neovascularizafion(NV). Methods Newborn Sprague-Dawley rats were randomly divided into acidosis-induced retinopathy group( ROP group) and control group. VEGF and Ang-2 in retina were detected by immunohistochemistry. Results The result of immunohistochemistry revealed that the protein expression of VEGF and Ang-2 of the ROP group on the 8th day was lower than that of the control group (P 〈0.05) and the protein expressions of VEGF and Ang-2 on the 10th and 13th day were stronger than those of the control group (P 〈 0.05). The changes of VEGF and Ang-2 protein expression were the same. Conclusion The periods of acidosis result in down-regulation of VEGF and Ang- 2. When acidosis stops, the expression of VEGF and Ang-2 is up-regulated. The abnormal retina VEGF and Ang-2 expression may play an important role in the development of neovascularization in ROP.
