槲皮素对TGF-β诱导的人肺癌A549细胞上皮间质转化的影响

摘　　要：目的探讨槲皮素对TGF-β诱导的人肺癌A549细胞上皮间质转化的影响。方法在体外分别采取槲皮素及阴性对照DMSO干预经或不经TGF-β(10ng/mL)诱导的人肺癌A549细胞12、24、48、72h后,MTT法检测不同浓度槲皮素对A549细胞增殖的影响;RT-PCR和Westem blot实验检测上皮间质转化标志性因子E-cad-herin、N-cadherin、Vimentin的表达变化。结果与阴性对照组相比,不同浓度槲皮素作用的实验组A549细胞的增殖能力降低,同时在mRNA和蛋白水平E-cadherin上调以及N-cadherin、Vimentin的表达下调(P<0.05)。结论槲皮素能够抑制TGF-β诱导的人肺癌A549细胞的增殖,且呈时间-剂量依赖性,阻止A549细胞发生上皮间质转化,降低A549细胞的迁移侵袭运动能力。Objective To investigate the effect of Quercetin on epithelial-mesenchymal transition in human lung ep- ithelial cell line A549 induced by TGF-β. Methods A549 cell was divided to 3 groups： no TGF-β group,DMSO control group,and TGF-β （10 ng/mL）＋ Quercetin group. After 12,24,48 and 72 h of treatment,proliferation of cell was determined with methabenzthiazuron（MTT） assay.The mRNA expression of E-cadherin,N-cadherin and Vimentin was determined by real-time RT-PCR. The proteins of E-cadherin,N-cadherin and Vimentin were determined by Western blotting. Results Compared with the control group,the proliferation of TGF-β （10 ng/mL）＋Quercetin group was significantly decreased. Meanwhile,E-cadherin was increased,N-cadherin and vimentin was obviously up-regu- lated at mRNA and protein level in A549 cells. Conclusion Quercetin can markedly inhibit the proliferation of lung adenocarcinoma cell line A549 induced by TGF-β in time-dependent and dose-dependent manners,prevent epithe- lial-interstitial transformation of A549 cells,decrease the migration and metastatic abilities of A549 cells.
