摘 要:目的探讨不同浓度茶多酚对体外培养人胚肝细胞(L-02细胞)生存率的影响。方法体外培养L-02细胞,采用50、100、250、500、1 000 mg/L浓度的茶多酚分别干预12、24、36、48 h,倒置显微镜观察各组细胞形态学的变化,用噻唑蓝法测定细胞的存活率。结果与对照组比较,250 mg/L及以上浓度茶多酚组干预12 h后细胞形态即发生变化,初期表现为细胞皱缩、体积变小,而后细胞固缩变为圆形,出现大量漂浮细胞及细胞碎片,形态学变化的程度与茶多酚浓度及作用时间有关。50 mg/L、100 mg/L茶多酚组作用48 h时L-02细胞存活率均高于对照组(P<0.05);250 mg/L及以上浓度茶多酚组干预后L-02细胞存活率均低于对照组(P<0.05),并且随着作用时间的延长和茶多酚浓度的增加,L-02细胞存活率减少(P<0.01)。结论低剂量(≤100 mg/L)茶多酚可能具有促进肝细胞生长、保护肝细胞的作用,较大剂量(≥250 mg/L)则有细胞毒性和抑制细胞生长的作用。Objective To explore the influence of tea polyphenols(TP) of different contents on the survival rate of L-02 cells cultured in vitro. Methods L-02 cells cultured in vitro,which were interfered by tea polyphenols of different concentrations (50,100,250,500 and 1000 rag/L) in four intervals of time respectively, i. e. 12,24,36 and 48 hours. Inverted microscope was applied to observe the cellular morphological changes, and the cell survival rate was determined by MTY assay. Results Compare with the control group, the cell morphological changes were observed in TP groups of 250 mg/L and above, which showed crenulation and shrinkage in cell volume in the early stage, then the cell pyknosis in round, plenty of floating cells and cell debris could be seen. The morphological change correlated with TP concentration and rip intervention time. Compare with the control group ,the higher survival rates of L-02 cells were observed in TP groups of 100 mg,/L and 50 rag/L, which were interfered for 48 hours (P 〈 0.05 ) ;After intervention, the survival rates of L-02 cells of rip groups of 250 mg/L and above were lower than that of the control group(P 〈0.05 ), which decreased with the prolonged time of drug action and increasing TP concentration ( P 〈 0. 01 ). Conclusion Low-dose tea polyphenols ( ≤ 100 mg/L) may be of protective effects on hepatoeytes and promote cell growth while high-dose one(250 mg/L) may be of cytotoxic effects and inhibition of cell growth.
