摘  要:本文应用RP-HPLC建立了小鼠肾脏中儿茶素EGCG检测方法,同时探索儿茶素EGCG在小鼠肾脏组织分布特征。小鼠腹腔注射后,不同时间点采集肾脏组织,RP-HPLC检测其中EGCG含量,计算儿茶素EGCG在肾脏中的分布。研究结果表明所建立的HPLC检测方法在0.5~200mg/L范围内线性关系良好(r>0.99),平均回收率85.0%以上,日内和日间精密度均低于10.0%。该方法操作简便、灵敏、准确,适合于肾脏组织分布研究。The study was designed to investigate the distribution of epigallocatechin gallate (EGCG) in mouse kidney, based on establishing a RP-HPLC method for determination of EGCG Kidney tissue was collected after administeration by intraperitoneal injection. Then EGCG concentrations were calculated by RP-HPLC method mentioned above. The results showed that there was a good linearity over the range 0.5-200.0 mg/L (r〉0.99). Then the recoveries were more than 85.0% in kidney samples, and the intra- and inter-day coefficients of variation were less than 10.0% in all cases. The RP-HPLC method was specific, simple, sensitive and accurate. And this method was successfully applied to EGCG distribution in mouse kidney.
