中低剂量四氯化碳联合食用乙醇作肝硬化造模

摘  要:目的探索四氯化碳(CCI4)联合食用乙醇进行大鼠肝硬化造模的可行性及剂量的把握.方法80只Wistar大鼠随机分为四组:CCI4组,食用乙醇组,CCI4加食用乙醇组,空白对照组.实验组分别以40%CCI4-橄榄油溶液3ml/kg腹腔注射(1次/4d)、45%食用乙醇灌胃(1ml·100g-1·d-1)加橄榄油腹腔注射(1次/4d)、40% CCl4-橄榄油溶液2.5mk/kg腹腔注射(1次/4d)加45%食用乙醇灌胃(1ml·100g-1·d-1),对照组仅腹腔注射同等体积橄榄油(1次/4d).第4、6、8周末随机取2只大鼠剖腹肝活检,观察成模情况,并检测肝功能.结果 CCI4加食用乙醇造模大鼠6周末即可见肝硬化结节,10周末明显肝硬化,成模率100%,死亡率17.39%;单用CCI4组8周末可见少量肝硬化结节,10周末明显肝硬化,成模率100%,死亡率30.43%;单用乙醇组10周末表现为肝脂肪变,无实验性死亡;对照组实验过程中肝组织无明显变化,无实验性死亡.结论中低剂量CCI4联合用食用乙醇肝硬化造模成模时间短,成模率高.Objective To study the feasibility and dosage of establishing hepatic cirrhosis rat models with CCI4 plus alcohol. Methods 80 Wistar rats were randomly divided into four groups:CCI4 group(group1),alcohol group(group2),CCI4 plus alcohol group(group3)and control group(group4). Experimental groups were given respectively intraperitoneal injection with 40%CCI4-olive oil mixture(3ml·kg-1)per four days,irrigation with 45%alcohol(1ml·100g-1·d-1)and intraperitoneal injection with olive oil every four days and intraperitoneal injection with 40%CCI4-olive oil mixture(2.5ml·kg-1)per four days,and intraperitoneal injection with 45%alcohol 1ml.100g-1·d-1,control group were administered with the same volume of olive oil every four days. At the end of 4th,6th and 8th week,two rats were selected randomly for liver biopsy and liver function test,which could be observe the situation of establishing hepatic cirrhosis rat models. Results Liver cirrhotic dodules could be seen in group3 at the end of 6th week and obvious iver cirrhosis at the end of 10th week with modeling rate of 100%and mortality rate of 17.39%;In contrast,little liver cirrhotic dodules could be seen in group1 at the end of 8th week and the similar Results at the end of 10th but with mortality rate of 17.39%;While only hepatic steatosis and no no experimental death in group2;no obvious changes and no experimental death in group4. Conclusion The establishment of hepatic cirrhosis rat models with lower dosage of CCI4 plus alcohol needs shorter time, which has a higher success rate of model establishment and a lower mordality.
