摘 要:目的探讨脂肪酸结合蛋白3(FABP3)过表达对斑马鱼胚胎活性氧(ROS)及线粒体ATP含量的影响。方法构建FABP3过表达载体,在斑马鱼受精卵的单细胞期进行胚胎显微注射(A组);以野生型斑马鱼作为空白对照(C组)。收集受精后24、48hpf斑马鱼胚胎,采用流式细胞术和化学发光法分别检测斑马鱼ROS和ATP含量。结果与C组相比,A组24hpf时斑马鱼ROS水平降低,ATP含量增加(P<0.05);而在48hpf时,两组间斑马鱼ROS水平和ATP含量无统计学差异(P>0.05)。结论 FABP3过表达对斑马鱼线粒体功能起保护作用。Objective To investigate the effect of fat acid binding protein 3(FABP3) overexpression on the levels of reactive oxygen species(ROS) and mitochondrial ATP in zebrafish embryo. Methods The vector overexpressing FABP3 was constructed and microinjected into the single cell stage of zebrafish zygote in group A, and the wild type of zebrafish embryo was taken as blank control (group C). After collecting the fertilized zebrafish embryos at 24 and 48 hpf,the contents of ROS and mitochondrial ATP were detected by flow cytometry and chemical luminescence method, respectively. Results Compared with group C, the level of ROS was decreased, while the content of ATP was increased at 24 hpf in group A(P〈0. 05). However, there was no significant difference in the levels of ROS and ATP at 48 hpf between groups of A and C(P〉0.05). Conclusion FABP3 overexpression can protect the mitochondrial function in zebrafish.
