摘  要:目的探讨Chibby在非小细胞肺癌(non-small celllung cancer,NSCLC)的表达及与临床病理因素,β-catenin表达之间的关系。方法采用免疫组织化学SP法检测100例NSCLC及相应癌旁正常肺组织中Chibby和β-catenin的表达,采用Western Blot方法检测35例新鲜肺癌组织及其癌旁正常肺组织中Chibby的表达情况。结果 Western Blot结果显示NSCLC中Chibby蛋白表达水平与癌旁正常肺组织相比无显著差异(>0.05)。免疫组化结果显示Chibby在NSCLC组织中的细胞核阳性率为43.0%(43/100),其在正常肺组织中的细胞核阳性率为82.0%(82/100),差异显著(<0.05);肺腺癌中Chibby细胞核阳性率57.9%(33/57)显著高于其在肺鳞癌中的阳性率23.3%(10/43,<0.05);无淋巴结转移组中Chibby的细胞膜阳性率38.9%(21/54)显著高于淋巴结转移组19.6%(9/46,<0.05)。β-catenin在细胞浆中的异常表达与NSCLC的分化程度呈负相关(<0.05)。Spearman相关性分析发现,NSCLC中Chibby在细胞膜/浆中的定位分别与β-catenin在细胞膜/浆的定位具有相关性(<0.05)。结论 Chibby在NSCLC组织中的细胞核表达显著低于正常肺组织,Chibby可能作为β-catenin的分子伴侣并调控β-catenin在细胞内的定位。Objective To investigate the expression and correlation of Chibby and 13 -catenin in non-small cell lung cancer(NSCLC) and analyze their relationships with the elinicopatholo^cal parameters of NSCLC. Methods The expressions of Chibby and B -eatenin were examined in 100 NSCLC tissues by immunohistochemistry and Chibby protein was also detected in 35 fresh NSCLC tissues and adjacent normal tissues by Western Blot. Results Western Blot showed that there were no significant difference for the expression levels of Chibby protein between NSCLCs and adjacent normal lung tissuesP〉0.05), and the immunohistochemistry showed that the nuclear expression rate of Chibby was reduced significantly in NSCLC 43.0% (43/100) than in normal lung tissues g2%(g2/100,P〈0.05). The nuclear expression rate of Chibby in lung adenocarcinoma 57.9%(33/57 ) was significant higher than that in lung squamous cell carcinoma 23.3%(10/43, P〈0.05). The membrane expression of Chibby was negatively correlated with lymphatic metastasis(p〈0.05). The cytoplasmic expression of B -eatenin was correlated with poor differentiation of NSCLC(P〈0.05). Spearman showed that the expression of Chibby in cytoplasm and membrane was correlated with cytoplasmic and membrane expression of B - catenin respectively(P〈0.05). Conclusion The nuclear expression of Chibby is significantly reduced in NSCLC, Chibby may concurrently express with 13 -catenin in NSCLC.
