鸡肝中8种磺胺类药物残留液相色谱检测方法的建立

摘　　要：目的:建立了同时测定鸡肝中磺胺嘧啶、磺胺甲基嘧啶、磺胺二甲氧嘧啶及磺胺对甲氧哒嗪等8种磺胺类药物的高效液相色谱检测方法,比较和优化了提取及色谱条件。方法:样品经乙腈提取,碱性氧化铝固相萃取柱净化后,洗脱液过0.45μm有机微孔滤膜,用高效液相色谱仪直接测量,外标法定量。采用XDB-C18色谱柱进行分离,以0.2%乙酸溶液和甲醇乙腈水混合液为流动相进行等度洗脱,紫外检测器检测,检测波长270 nm。结果:8种磺胺类药物标准曲线的线性相关系数大于0.997,线性范围0.1μg/kg～5μg/kg。该方法平均回收率为81.5%～91.2%,相对标准偏差小于12.7%。结论:该方法适合于鸡肝样品中上述8种磺胺类药物的检测,结果准确可靠。Objective：This study established a method for determination of sulfonamides residues in chicken by high performance liquid chromatography, comparison and optimization of extraction conditions were also done. Methods ： Samples were extracted with acetonitrile, purified with alkaline aluminum oxide column, using 0.45 μm organic microporous membrane to filter eluate, then measured by high performance liquid chromatography directly, quantified by external standard method. Samples were separated on XDB -C18 column, with 0.2% acetic acid so- lution and methanol acetonitrile/water mixture as the mobile phase for isocratic elution, UV detector was used for detection and the detection wave length was 270 nm. Results ： The eight kinds of sulfonamides showed good linearity in the range of 0.1 μg,/kg - 5 μg/kg（ r 〉 0. 997 ）. The average recovery rate was 81.5% - 91.2%. The relative standard deviation is less than 12.7%. Conclusion： The method is suitable for detection of 8 kinds of sulfonamides in chicken liver, with accurate and reliable results.
