摘 要:目的观察FIZZ1在慢性阻塞性肺疾病(COPD)大鼠肺组织中表达水平的动态变化,了解其在COPD发病中的作用机制。方法 40只雄性SD大鼠随机分为4组:正常对照组10只(A组),COPD模型2周组10只(B组),COPD模型4周组10只(C组),COPD模型八周组10只(D组)。常规病理切片观察各组大鼠气道和肺组织的形态学变化,免疫组织化学染色法检测肺组织中FIZZ1的蛋白水平,RT-PCR法检测肺组织中FIZZ1的mRNA的表达情况。结果 COPD大鼠被激发后2周开始出现气道壁及平滑肌增厚,气道上皮受损,部分纤毛脱落,气道周围及肺泡间隔内有大量炎症细胞浸润,且随着激发时间的延长,以上变化更加明显。同时,COPD大鼠支气管肺组织中FIZZ1的蛋白表达以及FIZZ1的mRNA表达水平逐渐升高,呈现一定的动态变化。结论 FIZZ1在COPD大鼠模型中呈动态升高过程,可能参与了COPD的发生发展。【Objective】 To observe the dynamic expression of FIZZ1 in the chronic obstructive pulmonary disease rats,understanding the mechanisms of its role in the pathogenesis of COPD.【Method】 40 healthy SD rats were randomly divided into four groups: the normal control group(group A) with 10,the COPD model group with 30(group B,divided into COPD model two weeks group,four weeks group C,eight weeks group D,10 in each group).The airway and lung tissue morphological change was observed by normal slice.Immunohistochemistry and RT-PCR were used to measure the level of FIZZ1 alpha protein and mRNA in the rat lung tissues.【Result】 COPD model two weeks of bronchial wall and airway smooth muscle thickened,airway epithelial damaged,part of the cilia fell off,there were a lot of inflammatory cells infiltrating surrounding the airways and alveolar interval,and stimulated the extension of time,the above changes were more obvious.Of COPD the rat bronchial lung tissue in FIZZ1 protein expression and FIZZ1 of mRNA expression gradually increased,showing the dynamic changes.【Conclusion】 The expression of FIZZ1 is a dynamic changing in the chronic obstructive pulmonary disease.FIZZ1 may be involved in the development process of COPD.
