SD大鼠缓解-复发型实验性变态反应性脑脊髓炎模型的建立

摘　　要：目的:建立大鼠缓解-复发型实验性变态反应性脑脊髓炎(EAE)模型,进行病理学研究,为多发性硬化(MS)的研究提供依据。方法:采用大鼠脊髓和弗氏完全佐剂混合乳剂一次性注入SD大鼠双足垫和尾部,1周后半剂量注射进行一次加强,诱导大鼠发生EAE;观察发病情况,HE染色观察病理变化,监牢兰染色观察脱髓鞘情况。结果:空白对照组大鼠均未出现症状,HE染色小脑及脊髓无炎性细胞浸润,监牢兰染色未见髓鞘脱失;模型组临床症状发生率为90%以上,HE染色可见小脑白质及脊髓血管周围有大量炎性细胞浸润,监牢兰染色发现有大片髓鞘脱落。结论:用SD大鼠脊髓髓鞘蛋白和弗氏完全佐剂混合乳剂可诱导同种SD大鼠发生缓解-复发型EAE。Objective： To investigate establishing the relapsing-remitting model aud pathological charaeteristit＇s of experimental allergic enCelphalomyelitis（EAE） model, and to provide the experimental value for researching muhi- pie s~.lerosis （MS）. Methods： The animal model were established in SD rats by immuuizatiou with eumplele Freund adjuvaut（Cli＇A） and GPSCH and boosted once. The clinical manifestation was obserw＊d. Inflammatory cell iufi]h＇atiou and demveliuation were detected by HE stain and fast blue stain respectively. Results： Over 80% rats dew,loped to EAE in group treated with GPSCH and CFA but none of the control groups. The intlammatiou cell were foul;d aroulld vessels aud demvelination in white matter of cerebellum and spinal eords. Conclusion： Relaps- ing-remitting EAE can be induced in SI） rats by immunization with the homogenate of GPSCtt and CFA.
