摘  要:目的:通过改良手术方式阻断肾动脉,观察肾缺血再灌注损伤(renal ischemia reperfusion injury,IRI)成模情况,为IRI模型建立提供新方法。方法:雄性SD大鼠42只,随机分为正常组(n=5)、假手术组(sham group,SG)(n=5)和模型组(n=32),模型组下设IRI后1h、6h、12h、24h4个时间点,每个时间点8只大鼠。手术方式:将大鼠麻醉后,逐层开腹暴露双肾,用玻璃分针分离双侧肾动脉,将已经消毒的压脉带片段包裹在肾动脉外层,将手术线系在压脉带片段外迅速打结阻断肾血流,观察肾脏颜色的变化。阻断肾血流45min后,剪断手术线,恢复肾脏供血,观察肾脏颜色变化。逐层关腹,复苏。观察IRI后尿量变化、肾功能和肾组织病理改变。结果:与正常对照组和SG组相比,IRI组尿量明显减少;IRI各组血肌酐(Scr)和尿素氮(BUN)升高明显(P<0.05);病理改变明显(P<0.05)。结论:通过简易手术结扎双侧肾动脉可以成功建立大鼠IRI损伤模型,成模率高,简单易行,成本效益高。Objective:Providing a novel method to construct the model of renal ischemia/reperfusion injury (IRI) ,Through improve the surgical approach of block the bilateral renal arteries of rat,the model of renal ischemia- reperfusion injury(IRI) into a mold rate were examined. Methods :A total of 42 male Sprague- Dawley rats were randomly divided into :normal group( n = 5 ) ,sham group( n = 5 ) and IRI group( n = 32). IRI group were divided into four time points : 1 hour,6 hour, 12 hour,24 hour, each time point contain 8 animals. Surgical Procedure : After the rats were anesthetized, midline laparotomy, both renal were exposed, bilateral renal blood flow were interrupted for 45min by applying rubber strips that wrapped in the vessels outside and ligation. Rubber strips were re- moved and blood flow restored at the end of 45min of ischemia. Observing the color changes of the kidneys in the whole operation process. Closed the belly, recovery. Renal function and pathology of the kidneys were examined. Results: Compared with the normal group and the sham group,urine output were remarkably decreased in IRI groups, Levels of serum creatinine(Scr) and blood urea ni- trogen(BUN) as well as histological scores were significantly increased in IRI groups. Conclusion:Results show that stable model of renal IRI can be gained in rat by a simple surgical to incarcerate bilateral renal arteries. The successful rate of modal was very high,It not only a cost -efficient but also a simple and practicable method to construct the model.
