转化生长因子β1对A549细胞晚期糖基化终产物受体与细胞外基质表达的影响

摘  要:目的研究转化生长因子Bl(transforminggrowthfactorbeta1,TGF-β1)对人A549细胞的晚期糖基化终产物受体(receptorforadvancedglycationend—products,RAGE)和胶原-I、a-平滑肌肌动蛋白(a-smoothmuscleactin,d—SMA)表达的作用。方法体外培养人A549细胞,以不加TGF-[31刺激为对照组,不同终浓度TGF-β1(2μg/L、5,ag/L、10/μg/L)处理A549细胞为实验组,RT-PCR和Westernblotting法检测TGF-β1刺激12h、24h、36h后对照组和实验组RAGEmRNA和RAGE、胶原-I及a—SMA的蛋白表达。结果①随着TGF-β1,浓度的增加,刺激时间的延长,A549细胞的RAGEmRNA和蛋白的表达逐渐降低,呈时间-浓度依赖关系。②与对照组相比,A549在TGF-β1(5μg/L)刺激24h后,RAGEmRNA的表达明显降低(0.387±0.088VS1.345±0.132,P〈0.01)。RAGE蛋白表达较对照组明显降低(O.174±0.061VS1.229±0.112,P〈0.01)。③TGF-β1(5μg/L)刺激24h后,A549的胶原-I及d—SMA蛋白表达(0.916±0.071和0.899±0.022)分别较对照组(0.295±0.041和0.134±0.028)升高3.1和6.7倍,差异有统计学意义(P〈O.01)。④相关分析显示,A549的RAGE蛋白表达与胶原-I(r=-0.843,P〈0.05)、n—SMA蛋白的表达(r=-0.897,P〈0.05)呈负相关。结论TGF-β3。下调A549细胞RAGE的mRNA和蛋白表达,与胶原-I及a—SMA蛋白表达上调呈负相关,提示RAGE的表达下调可能与肺纤维化的发生和发展有关。Objective To investigate the expressions of receptor for advanced glycation endproducts (RAGE), collagen-I , and asmooth muscle actin (a-SMA) in human A549 cells induced by transforming growth factor beta 1 (TGF-β1 ). Methods With no TGF-β1-stimulated cells for the control group, human A549 cells were cultured in vitro. Cultured cells were exposed to TGF-β1with different final concentration (2 /μg/L,5 g/L,10 μg/L) for different time (12 h,24 h,36 h). RAGE mRNA and protein, collagen- I and a-SMA in cells were detected at different points after the treatment of by RT-PCR and Western blotting. Results @TGF-β1reduced RAGE mRNA and protein expressions in a dose- and time-dependent manner. @Compared with the control group, RAGE mRNA (0. 387±0. 088 vs 1. 345±0. 132) and protein expressions (0. 174±0. 061 vs 1. 229±0. 112) were significantly reduced ( P 〈 0.01) in cells exposed to TGF-β1 (5 /μg/L) for 24 hours. @The collagenI and a-SMA protein expressions increased after stimulation for 24 hours in 5 μg/L TGF-β1 group. At that point, collagen- β1 and a-SMA protein expressions were 3.1 and 6.7 times of the control group ( P 〈0.01). @RAGE expression had a negative correlation with collagen-I (r =--0. 843, P 〈0.05) and a-SMA protein expression (r = -0. 897, P 〈0.05). Conclusions The reduced RAGE, which is negatively correlated with expressions of collagen- I and a-SMA proteins in A549 cells exposed to TGF-β1, may be related to the development and progress of pulmonary fibrosis.
