摘 要:目的研究铁对体外培养的人肺腺癌细胞SPC-A-1及肿瘤血管内皮细胞模型增殖活性的影响。方法采用四甲基偶氮唑盐(MTT)法检测人肺腺癌细胞SPC-A-1及肿瘤血管内皮细胞模型的增殖活性,流式细胞仪检测铁对SPC-A-1细胞周期和细胞凋亡的效应。结果 10、30、100及300 mol/L浓度组铁可明显促进SPC-A-1细胞的增殖活性;10、30及100 mol/L浓度组铁使SPC-A-1的G1期细胞比例减少,S期和G2期细胞比例增多;30及100 mol/L浓度组铁使SPC-A-1的凋亡率显著降低;30及100 mol/L的铁可明显促进肿瘤血管内皮细胞的增殖活性。结论铁可通过促进肿瘤细胞向S期转化、降低凋亡率从而促进SPC-A-1细胞增殖,还可通过刺激血管内皮细胞的增殖促进肿瘤的增长。Objective To study the effect of iron on the proliferation of human lung adenocarcinoma cell SPC-A- 1 and tumor vascular endothelial cells. Methods Proliferation activity of SPC-A-1 and tumor vascular endothelial cells was monitored by MTT assay. Flow cytometry was performed to measure the cell cycle and apoptosis. Results Iron at 101μmol/L, 30μmol/L, 1001μmol/L, 3001μmol/L concentration significantly increased proliferation of SPC- A-1. Iron at 101μmol/L, 301μmol/L, 100 1μmol/L concentration decreased the cell counts in G1 phase while it increased the cell counts in S phase and G2 phase. Iron at 30 1μmol/L, 100 1μmol/L concentration reduced the ratio of apoptosis of SPC-A-1. The proliferation of tumor vascular endothelial cells were obviously increased in iron groups at 30 1μmol/L, 1001μmol/L concentration. Conclusions Iron could increase the proliferation of SPC-A- 1 by prompting the cells en- tering S phase and reducing the ratio of cell apoptosis. Iron could also increase the growth of tumor through promoting the proliferation of tumor vascular endothelial cells.
