摘  要:目的初步研究氢饱和生理盐水对SD大鼠亚硒酸钠性白内障的抑制作用。方法选用7日龄健康SD大鼠68只,随机分为6组,正常对照组、白内障组、氢水预防组、氢水早期治疗组、氢水晚期治疗组和氢水组。白内障组、氢水预防组、氢水早期治疗组和氢水晚期治疗组大鼠12日龄按25μmol.kg-1体质量单次颈背部皮下注射2mmol.L-1亚硒酸钠生理盐水。氢水预防组大鼠8~12日龄腹腔注射氢饱和生理盐水5mL.kg-1,每天1次,连续5d;氢水早期治疗组大鼠12~17日龄,注射处理相同;氢水晚期治疗组大鼠17~21日龄;氢水组大鼠8~17日龄进行注射。于26日龄裂隙灯显微镜下观察大鼠晶状体混浊程度,测定晶状体中SOD活性及GSH、MDA含量。结果正常对照组、氢水组晶状体均透明,白内障组、氢水预防组和氢水晚期治疗组晶状体完全混浊;氢水早期治疗组晶状体混浊程度明显低于白内障组,差异具有统计学意义(χ2=30.48,P<0.05)。氢水早期治疗组与白内障组比较,晶状体中SOD活性及GSH含量明显升高(SOD:q=13.13,P<0.01;GSH:q=11.76,P<0.01),但白内障组和氢水预防组(SOD:q=1.96,P>0.05;GSH:q=1.61,P>0.05;MDA:q=2.74,P>0.05)及氢水晚期治疗组(SOD:q=1.37,P>0.05;GSH:q=1.25,P>0.05;MDA:q=1.11,P>0.05)之间比较差异均无统计学意义。氢水早期治疗组MDA含量升高不明显,明显低于白内障组(q=10.28,P<0.01)。结论氢饱和生理盐水可以提高晶状体组织中SOD活性及GSH含量,减少MDA生成,从而减轻晶状体的氧化损伤,对亚硒酸钠性白内障的发生和发展有一定的抑制和延缓作用,但无明显的预防作用,对于已形成的白内障无逆转作用,对晶状体无明显的毒性作用。Objective To study prelim inarily the inhibitive effect of saturated hydrogen saline on sodium selenite-induced cataract in SD rats.M ethod s At the1st day,68 7-day-old healthy SD rats were random ly divided into6groups:normal control group,cataract group,Saturated hydrogen saline prevention group(the H2 saline prevention group),the early Saturated hydrogen saline treatment group(the early H2 saline treatment group),the late Saturated hydrogen saline treatment group(the late H2 saline treatment group)and the Saturated hydrogen saline group(the H2 saline group).At the 6th day,rats in the cataract group,the H2 saline prevention group,the early H2 saline treatment group and the late H2 saline treatment group were subcutaneously injected w ith a single dose(25μmol.kg-1)of sodium selenite saline(2mmol.L-1).Intraperitoneal injection of saturated hydrogen saline(5mL.kg-1)was performed on rats in the H2 saline prevention group from the 2nd to the6th day,rats in the early H2 saline treatment group from the6th to the11th day,rats in the late H2 saline treatment group from the11th to the16th day and rats in the H2 saline group from the 2nd to the11th day,one time per day.At the 2 0th day,crystal lenses opacification in rats were observed by slitlamp exam ination.superoxide dismutase(SOD) activity and levels of reduced glutath-i one(GSH)and malondialdehyde(MDA) in rat crystal lens were also detected.R esu lts A ll crystal lens were normal in the normal control group and the H2 saline group.Crystal lens in the cataract group,the H2 saline prevention group and the late H2 saline treatment group were totally opacified.crystal lens opacification level in the early H2 saline treatment group is obviously lower than that in the cataract group,the difference was of statistical significance(χ2 =30.48,P0.05).SOD activity and levels of GSH and MDA in crystal lens of rats in the early H2 saline treatment group were significantly higher than that in the cataract group(SOD:q=13.13,P0.01;
