玉屏风散对单纯疱疹病毒性角膜炎模型小鼠细胞免疫功能的影响

摘　　要：目的观察中药玉屏风散对不同感染时期单纯疱疹病毒性角膜炎(herpes simplex keratitis,HSK)小鼠模型细胞免疫水平的影响,探讨其抗HSK复发的作用及其机制。方法采用流式细胞术分析不同感染时期模型小鼠脾脏+、+、+淋巴细胞表达水平,ELISA法检测血清γ-IFN、IL-2、IL-4及IL-10水平改变。观察并比较玉屏风散对不同感染时期HSK小鼠模型CD3+、CD4+、CD8+T淋巴细胞和血清γ-IFN、IL-2、IL-4及IL-10水平改变的影响。结果与正常对照组比较,HSK潜伏期(PI28d)和复发期(UVB3d)模型小鼠CD3+、CD4+T淋巴细胞表达明显降低,CD8+T淋巴细胞表达明显升高;外周血Th1型细胞因子IL-2、γ-IFN水平显著降低,Th2型细胞因子IL-4、IL-10水平显著增高。CD4+T淋巴细胞表达的降低与IL-2、γ-IFN水平的降低呈显著正相关。在同一感染期内,玉屏风散治疗组、玉屏风散+阿昔洛韦治疗组治疗末期与治疗开始时比较,HSK模型小鼠体内CD3+、CD4+、CD8+T淋巴细胞表达水平和外周血Th1型细胞因子IL-2、γ-IFN,Th2型细胞因子IL-4、IL-10表达水平均有明显改善,并显著优于单纯阿昔洛韦治疗组及空白对照组。结论玉屏风散具有提高HSK潜伏期和复发期模型小鼠体内CD3+、CD4+T淋巴细胞表达,降低CD8+T淋巴细胞表达,改善CD4+/CD8+比值水平,调节机体细胞免疫功能的作用。Objective To investigate the anti-recurrence effect of Yu Ping Feng Powder on herpes simplex keratitis（HSK）and the mechanism of that by observing its influence on cellular immune function of HSK model mice at different infection stage.Methods The expression level of CD3＋,CD4＋,CD8＋（T-lymphocytes）at different infection phases were detected by flow cytometry.The content of γ-IFN,IL-2,IL-4 and IL-10（cytokines）in serum at different infection phases were also tested by ELISA.Results of those two tests were analyzed.Results The CD3＋ and CD4＋ expression level at HSK latency stage（PI 28 days）and recurrence stage（UV-B 3 days）were significantly lower,while CD8＋ expression level were significantly higher,comparing with those in the normal control group.The γ-IFN,IL-2（Th1-type）content in peripheral blood（PB）at HSK latency stage（PI 28 days）and recurrence stage（UVB 3 days）were significantly lower,while IL-4 and IL-10（Th2-type）content were significantly higher,comparing with those in the normal control group.At the final phase of the treatment,the CD3＋,CD4＋,CD8＋ expression level and PBγ-IFN,IL-2,IL-4 and IL-10 content were greatly improved,comparing with the beginning phase of the treatment,both in the Yu Ping Feng Powder treatment group and the Yu Ping Feng Powder-acyclovir treatment group at the same infection stage.The CD3＋,CD4＋,CD8＋ expression level and PBγ-IFN,IL-2,IL-4 and IL-10 content in the Yu Ping Feng Powder treatment group and the Yu Ping Feng Powder-acyclovir treatment group were obviously improved than those in the acyclovir-only treatment group and the blank control group.Conclusion Yu Ping Feng Powder can enhance the CD3＋ and CD4＋ expression,weaken the CD8＋ expression and improve the value of CD4＋/CD8＋,which can strengthen cellular immune function.
