摘  要:目的检测microRNA-155(miR-155)在胰腺导管腺癌(pancreatic ductal adenocarcinoma,PDAC)组织中、患者血清中的含量,探讨其作为PDAC血清学诊断标志物的可能性。方法应用荧光实时定量PCR法检测miR-155在20对PDAC组织和癌旁组织、70例PDAC患者和40例健康人血清、10对荷人PDAC小鼠和对照组小鼠血清及6株PDAC细胞株和其培养上清中的含量。结果实验结果显示,PDAC组织中miR-155表达高于癌旁胰腺组织(P=0.005);并且miR-155在PDAC患者血清中表达量高于健康对照组(P=0.004);在荷人PDAC小鼠血清中miR-155含量高于对照组小鼠(P=0.012);同时,6株PDAC细胞株miR-155的表达存在差异(P=0.000),miR-155在细胞内和在其培养上清中的表达具有高度一致性(r=0.628,P=0.005)。结论 miR-155在PDAC组织和患者血清中表达上调,提示其可能是PDAC血清学的潜在标志物。Purpose To investigate the expression of microRNA-155 (miR-155) in pancreatic ductal adenocarcinoma (PDAC) tissue and serum, as well as diagnostic value of miR-155 in serum of PDAC. Methods Tissue specimens from PDAC and pancreas ( n = 20 and n = 20, respectively) , the sera of patients with PDAC and control group (n = 70 and n = 40, respectively) , the sera of SCID mice with humans' PDAC and control group (n = 6 and n = 6, respectively) , the 6 PDAC cells and supernatant were analyzed via real-time RT-PCR for miR-155. Results The expression of miR-155 in PDAC tissue was significantly higher than that in pancreatic tissue ( P = 0. 005 ). The expression of miR-155 in the sera of patients with PDAC was significantly higher than that in control group ( P = 0. 004). The expression of miR-155 in the sera of mice with humans' PDAC was significantly higher than that in control group (P = 0. 012). It was been found different expression level of miR-155 among 6 pancreatic cell lines (P = 0. 000), a positive correlation was observed between the expression of miR-155 in the 6 PDAC ceils and supernatant ( r = 0. 628, P = 0. 005 ). Conclusion The expression of miR-155 in the tissue and sera of patients with PDAC is significantly higher than that in control group. Therefore, serum miR-155 may be used as a potential biomarker in the diagnoses of PDAC.
