摘　　要：目的了解转铁蛋白受体(transferrin receptor,TfR)在皮肤黑素瘤中的水平及其临床意义。方法采用免疫组化法检测34例皮肤黑素瘤和20例色素痣石蜡标本中TfR的水平,流式细胞仪(FCM)检测TfR在黑素瘤细胞系A375,A875和KZ28细胞株的表达,并以文献报道的TfR阴性表达的人卵巢癌细胞系A2780作为阴性对照。结果在黑素瘤组织中,TfR的水平较色素痣对照组的水平明显增加,差异有统计学意义(P<0.05),并且TfR的水平与黑素瘤的病理分期、clark分级和有丝分裂均具有相关性;在黑素瘤细胞水平上,TfR在黑素瘤细胞系均高水平,且以A375水平最高。结论 TfR在皮肤黑素瘤中的水平明显增高,这为未来使用TfR作为基因治疗黑素瘤的靶向分子提供了坚实理论依据。Objective To study the significance of transferrin receptor（TfR） expression in malignant melanoma（MM） of human skin. Methods Expressions of TfR were detected in 34 cases of MM and 20 cases of pigmented nevus by immunohistochemical staining, and detected in melanoma cell line A375, A875 and KZ28 by immunohistochemistry techniques and Flow Cytometry, compared to human ovarian cancer cell line A2780 which were reported had no TfR expression. Results The expressions of TfR in malignant melanoma were significantly higer than that in pigmented nevus, and the level were related to pathological stage, clark level and mitotic rate. On cell level, all melanoma cell lines had high TfR expression, and A375 had the highest expression. Conclusion Expressions of TfR were significantly increased in malignant melanoma, which provided theoretical basis for the study of gene therapy of malignant melanoma.
