摘 要:目的:检测MicroRNA-21(miR-21)在结肠癌及其相应癌旁正常组织中的表达,探讨其与结肠癌临床病理指标的关系。方法:应用实时荧光定量PCR(RT-qPCR)技术检测50例结肠癌组织标本和癌旁组织标本中miR-21的相对表达量,分析其在2种组织中的表达变化,探讨miR-21表达水平与结肠癌发病位置、临床分期、浸润深度及有无淋巴结转移的关联性。结果:miR-21在结肠癌组织中相对表达水平显著高于癌旁组织(P<0.05),右半结肠癌组织中miR-21表达水平明显高于左半结肠癌组织(P<0.05);Ⅲ期和Ⅳ期结肠癌组织中miR-21表达水平明显高于Ⅰ期和Ⅱ期结肠癌(P<0.05);miR-21表达水平在不同浸润深度结肠癌之间比较差异无统计学意义(P>0.05);有淋巴结转移结肠癌组织中miR-21表达水平明显高于无淋巴结转移者(P<0.05)。结论:相对于癌旁组织,miR-21在结肠癌组织中表达明显上调,且表达水平与发病位置、临床分期及是否有淋巴结转移有关。Objective To detect the expression levels of MicroRNA-21(miR-21) in the colon cancer tissue samples and the cancer-adjacent tissues,and to explore the relationship between the clinicopathological features and the expression of miR-21.Methods Real-time florescence quantitative PCR(RT-qPCR) was used to detect the relative expression levels of miR-21 in 50 samples including the colon cancer tissue samples and the cancer-adjacent tissues;and the expression changes of miR-21 in two kinds of tissues were analyzed.The relationships between the miR-21 expression levels and the location,depth of invasion and lymph node metastasis of colon cancer were studied.Results The miR-21 relative expression level in colon cancer tissue samples was significantly higher than that in cancer-adjacent tissues(P0.05);the miR-21 expression level in right colon cancer tissues was obviously higher than that in left colon cancer tissues(P0.05);the miR-21 expression level in Ⅲ stage and Ⅳ stage colon cancer tissues was obviously higher than that in Ⅰ stage and Ⅱ stage colon cancer tissues(P 0.05);the expression levels of MiR-21 in colon cancer with different depths of invasion had no statistically significant differences(P0.05);the expression level of miR-21 in colon cancer tissues with lymph node metastasis was significantly higher than that in colon cancer tissues without lymph node metastasis(P0.05).Conclusion The miR-21 expression level is increased significantly in colon cancer tissues compared with cancer-adjacent tissues,and it is related to the location,the clinical stage and the lymphy node metastasis of colon cancer.
