扶正抗癌方联合吉非替尼治疗晚期非小细胞肺癌患者血浆差异蛋白比较分析

摘　　要：目的通过比较扶正抗癌方联合吉非替尼治疗晚期非小细胞肺癌患者1个月前后血浆蛋白,找出差异蛋白,为扶正抗癌方联合吉非替尼治疗非小细胞肺癌进一步研究提供差异蛋白组学证据基础。方法收集8例扶正抗癌方联合吉非替尼治疗的晚期非小细胞肺癌患者治疗前及治疗1个月后血浆,采用二维凝胶电泳(2-DE)技术分离血浆蛋白质,PDQuest 7.1.0图像分析软件识别差异蛋白点,基质辅助激光解析电离飞行时间质谱及液相色谱-电喷雾串联质谱技术鉴定差异蛋白。结果图像分析软件识别出11个差异蛋白点。基质辅助激光解析电离飞行时间质谱鉴定7种差异蛋白(异构体2的纤维蛋白原α链、载脂蛋白E、簇蛋白亚型2、甲状腺素、未知功能蛋白、皮质类固醇结合球蛋白、载脂蛋白A 4),未鉴定出的蛋白用液相色谱-电喷雾串联质谱技术鉴定出为Ⅱ型细胞骨架蛋白1。结论扶正抗癌方联合吉非替尼治疗晚期非小细胞肺癌对患者血浆蛋白有一定的影响,液相色谱—电喷雾串联质谱技术可作为基质辅助激光解析电离飞行时间质谱的补充使用。Objective It is to fine out differential protein to provide evidence basis of differential proteomics for the therapy of non-small cell lung cancer treated with Kangai Fuzheng decoction and gefitinib by comparing the plasma protein in patients with non-small cell lung cancer before treatment and one month after treatment with Kangai Fuzhcng decoction and gefitinib. Methods Plasma of 8 cases of patients with non-small cell lung cancer before treatment and one month after treatment with Kangai Fuzheng decoction and gefitinib was collected, and the plasma protein was separated by 2 - DE method, differential protein points were identified by PDQuest7.1.0 image analysis soft, differential protein was identified by MALDI- TOF- MS and UPLC - MS/MS. Results Image analysis soft found out 1 l differential protein points. MALDI - TOF - MS identified 7 kinds of differential protein（ Chain isomer of fibrinogen alpha 2 subtype protein apolipoprotein E cluster 2 thyroxine binding globulin （SHBG） with unknown function protein steroids apolipoprotein A4） , and the protein un-idcntified is Cytoskeleton protein 1 identified by UPLC - MS/MS. Conclusion Fuzheng Kangai decoction combined with gefitinib has some effect on the plasma protein in the patients with advanced non-small cell lung cancer, UPLC - MS/MS can be used as the supplement of MALDI - TOF.
