白细胞介素-4和肿瘤坏死因子-α在家兔油酸性急性肺损伤中的作用

摘　　要：目的探讨白细胞介素-4(IL-4)和肿瘤坏死因子-α(TNF-α)在家兔油酸性急性肺损伤(ALI)发病机制中的作用。方法 30只健康新西兰家兔随机分为油酸性ALI模型组(模型组)、IL-4干预组和对照组,每组10只,分别检测3组家兔动脉血氧分压(PaO2)及肺组织、外周血和支气管肺泡灌洗液(BALF)中TNF-α的表达水平,并进行比较。结果 IL-4干预组和模型组家兔PaO2显著低于对照组(P<0.05),IL-4干预组家兔PaO2显著高于模型组(P<0.05)。IL-4干预组和模型组家兔肺组织中TNF-α的表达显著强于对照组(P<0.05),IL-4干预组家兔肺组织中TNF-α水平显著弱于模型组(P<0.05)。模型组和IL-4干预组家兔外周血和BALF中TNF-α水平显著高于对照组(P<0.05),IL-4干预组家兔外周血和BALF中TNF-α水平显著低于模型组(P<0.05)。结论 IL-4和TNF-α可能参与家兔油酸性ALI的发生和发展。Objective To explore the effect of interleukin-4（IL-4）and tumor necrosis factor-α（TNF-α） on rabbits with acute lung injury（ALI）induced by oleic acid.Methods Thirty healthy New Zealand rabbits were randomly divided into oleic acid induced ALI model group（model group）,IL-4 treatment group and control group,with ten rabbits in each group.The levels of partial pressure of oxygen in artery（PaO2）and TNF-α in lung tissues,peripheral blood and bronchoalveolar lavage fluid（BALF）were detected and compared in the three groups.Results The levels of PaO2 in model group and IL-4 treatment group were significantly lower than those in control group（P〈0.05）,and the levels of PaO2 in IL-4 treatment group were significantly higher than those in model group（P〈0.05）.TNF-α levels in lung tissues of rabbits in model group and IL-4 treatment group were significantly higher than those in control group（P〈0.05）,and TNF-α levels in lung tissues of rabbits in IL-4 treatment group were significantly lower than those in model group（P〈0.05）.TNF-α levels in peripheral blood and BALF of rabbits in model group and IL-4 treatment group were significantly higher than those in control group（P〈0.05）,and TNF-α levels in peripheral blood and BALF of rabbits in IL-4 treatment group were significantly lower than those in model group（P〈0.05）.Conclusion IL-4 and TNF-α may play important roles in the occurrence and development of ALI induced by oleic acid in rabbits.
