摘 要:目的筛选长萼鸡眼草的高抗氧化活性萃取部,测定其总酚和总黄酮的含量。方法长萼鸡眼草乙醇提取物分别用石油醚、氯仿、乙酸乙酯、正丁醇萃取,利用清除OH.、O2-.和DPPH.实验以及还原能力等方法评价其不同极性萃取部的抗氧化能力。采用Folin-Ciocalteu法测其总酚含量,NaNO2-Al(NO3)3-NaOH显色法测总黄酮含量。结果长萼鸡眼草正丁醇萃取部和乙酸乙酯萃取部的抗氧化活性最强,对OH.和O2-.的清除效果显著高于VitC(P<0.05),石油醚萃取部和氯仿萃取部的抗氧化作用较弱。长萼鸡眼草正丁醇萃取部和乙酸乙酯萃取部的总酚含量分别为105.81±0.25、144.63±0.54 mg.g-1,总黄酮含量分别为199.15±2.63、396.24±5.13 mg.g-1。结论长萼鸡眼草的高抗氧化萃取部为正丁醇萃取部和乙酸乙酯萃取部,其抗氧化活性组分可能为酚酸类和黄酮类化合物,可作为天然抗氧化物质的来源。OBJECTIVE To screen the fractions with high antioxidant activity from the extracts of Kummerowia stipulacea and determine the total phenolic and flavonoid amours of the fractions. METHODS The ethanol extracts of K. stipulacea were fractioned by extration using petroleum ether, chloroform, ethyl acetate and n - butyl alcohol. Hydroxyl radical (OH·) , superoxide anion (O29), 1, 1-Diphenyl-2- picrylhydrazyl(DPPH) radical scavenging assays and reducing activity test were used to evaluate their antioxidant activity of the four fractions. Total phenolic contents were determined by Folin - Ciocaheu method, and total flavonoid contents were determined by NaNO2 - A1(NO3)3 - NaOH method. RESULTS Ethyl acetate fraction and n - butyl alcohol fraction exhibited the highest antioxidant activity. Their hydroxyl radical and superoxide anion scavenging activity were significantly higher than the control ascorbic acid(Vit C)(P〈0.05). The antioxidant activity of petroleum ether fraction and chloroform fraction was lower than that of ethyl acetate fraction and n - butyl alcohol fraction. Total phenolic contents of n - butyl alcohol and ethyl acetate fraction were 105.81±0.25 nlg.g^-1 and 144.63 ±0.54 mg.g^-1 ,respectively. Total flavonoid contents of n- butyl alcohol and ethyl acetate fraction were 199.15 ± 2.63 mg·g^-1 and 396.24±5.13 mg·g^-1, respectively. CONCLUSION The strong antioxidant activity fractions of K. stipulacea were n - butyl alcohol and ethyl acetate fractions. The most possible antioxidant phytochemicals were phenolics and flavonoids. K. stipulacea could be a source of natural antioxidants.
