摘  要:β-葡萄糖苷酶(β-glucosidase,BG)是昆虫降解纤维素起关键作用的限速酶,在糖原降解和糖类代谢方面具有重要的生理功能。本研究用粗纤维含量不同的饲料饲养家蝇Musca domestica,分为对照组(粗纤维含量为30%)、实验组1(粗纤维含量为40%)和实验组2(粗纤维含量为50%)。分别在家蝇发育的不同时期取样,采用实时荧光定量PCR技术和DNSA法检测β-葡萄糖苷酶基因的mRNA表达水平和β-葡萄糖苷酶活性。结果显示,β-葡萄糖苷酶基因mRNA表达水平和酶活性随家蝇的个体发育逐渐提高(P<0.05),并且与饲料中粗纤维的含量呈正相关(P<0.05)。实验组1β-葡萄糖苷酶基因mRNA表达水平较对照组提高了1.7~2.4倍,酶活性提高了1.04~1.37倍;实验组2mRNA表达水平较对照组提高了3.1~5.0倍,酶活性提高了1.19~2.25倍。这些结果表明取食粗纤维对家蝇β-葡萄糖苷酶基因表达量及酶活性具有诱导作用。β- glucosidase (BG) is a kind of critical enzyme for degrading cellulose in insects. It has an important function on glycogen degradation and carbohydrate metabolism. In this study, Musca domestica have been fed with crude fibre of different contents, which are divided into the control group ( the level of crude fiber is 30% ), the experiment group 1 ( the level of crude fiber is 40% ) and the experiment group 2 ( the level of crude fiber is 50% ). Real - time quantitative PCR was performed for detecting the expression of BG mRNA and DNSA method is used to detect the BG activity during the different development stage of M. domestica. And the experiment result shows there are significant difference of BG activity and its mRNA expression at every development stage, and moreover the BG activity and the mRNA expression increased with it's individual development. The BG activity and the mRNA expression keep in a significantly higher level when the content of crude fibre is enhanced for feeding musca domestica. Compared with the control group, the expression level of Musca domestica β- glycosidase gene and enzyme activity respectively increase by 1.7 to 2.4 times and by 1.04 to 1.37 times in experimental group 1, which increase by 3.1 to 5.0 times and by 1.19 to 2.25 times in experimental group 2. These result indicate that feeding M. domestica with crude fiber plays an inducing rolein the M. domestica β-glycosidase gene expression and enzyme activity.
