环氧合酶-2和HER-2/neu蛋白在卵巢上皮性癌中的表达及其临床意义

摘　　要：目的探讨环氧合酶-2(cyclooxygenase-2,COX-2)和表皮生长因子-2(v-erb-b2erythroblastic leukemia viral oncogene homolog2,HER-2/neu)蛋白在卵巢上皮性癌中的表达及其临床意义。方法利用免疫组化链酶素抗生物蛋白—过氧化物酶连接(streptavidin-perosidase,SP)法检测12例正常卵巢组织、18例卵巢良性上皮性肿瘤、10例卵巢交界性上皮性肿瘤和40例卵巢上皮性癌组织中COX-2和HER-2/neu蛋白的表达水平,并结合临床病理特征进行分析。结果①COX-2和HER-2/neu蛋白在卵巢正常组织、良性肿瘤、交界性肿瘤和上皮性癌中的阳性表达率分别为0%、5.6%、30.0%、85.O%和8.3%、11.1%、20.0%、52.5%,二者的表达率随肿瘤恶性程度增加而增加,差异有统计学意义(P<0.05)。②COX-2和HER-2/neu蛋白在卵巢上皮性癌中的表达均与临床分期、病理分级、组织学类型无关(P>0.05),与淋巴结转移有关(P<0.05),二者在卵巢上皮性癌组织中同时表达的阳性率为52.5%,呈正相关(r=0.44,P<0.05)。结论 COX-2和HER-2/neu蛋白与卵巢上皮性癌的发生发展密切相关,二者可能协同参与了卵巢癌的发生发展。Objective To study the expression and clinical significance of cyclooxygenase -2 （COX -2） and HER - 2/neu protein in ovarian epithelial cancer. Methods The expression of COX - 2 and HER - 2/neu protein was detected in 12 cases with normal ovarian tissues, 18 cases with benign ovarian epithelial tumor, 10 cases with borderline ovarian epithelial tumor and 40 cases with ovarian epithelial cancer by immunohistochemical technology streptavidin - perosidase（sp） method. Results The positive rates of COX - 2 and HER - 2/neu protein in normal ovary, benign epithelial ovarian tumor, borderline epithelial tumor, and malignant epithelial ovarian cancer tissue were 0% and 8.3% ,5.6% and 11.1% ,30. 0% and 20. 0% ,85.0% and 52. 5% respectively. With the increasing of the malignant degree of ovarian tumor, the positive rates of COX - 2 and HER - 2/neu protein both increased and the difference was significant（ P 〈 0.05 ）. Their expression was not correlated with clinical stage, histological type and pathology grade （ P 〉 0. 05 ）, but associated with lymph node metastasis （ P 〈 0. 05 ）. The positive rate of COX - 2 and HER -2/neu protein in the same specimens of ovarian cancer was 52. 5% , there was significant correlation between the expression of COX -2 and HER -2/neu protein （r =0. 44,P 〈0. 05）. Corldusion The overexpression of COX -2 and HER-2/neu may play an important role in the development of ovarian epithelial cancer,they may collaborative participation in the development of ovarian cancer.
