索拉非尼联合维生素K_2对人肝癌细胞裸鼠转移瘤模型的协同治疗效应

摘　　要：目的利用小鼠人肝癌细胞转移瘤模型,探讨索拉非尼联合维生素K2的协同治疗效应。方法构建小鼠人肝癌细胞转移瘤模型,分别用1.25mg/kg的索拉非尼和2mg/kg的维生素K2单独或联合给药,观察用药后小鼠肝内、肺内转移瘤情况及生存时间的变化。结果索拉非尼和维生素K2联合作用较空白对照及单独作用能够显著抑制小鼠肝、肺内转移灶的数量及体积,维持给药可显著改善小鼠生存状态并延长生存时间。结论索拉非尼和维生素K2对人肝细胞癌有良好的协同治疗效应,可抑制肿瘤肝、肺转移并延长生存期。Objective To investigate the synergistic therapeutic effects of sorafenib plus vitamin K2 on metastatic human hepatocellular carcinoma nude mice models.Methods After the establishment of metastatic human hepatocellular carcinoma models,the nude mice were treated with sorafenib（1.25 mg/kg） and vitamin K2（2 mg/kg） either alone or in combination.The development of tumor metastases in the liver or the lung and the survival time were observed.Results Compared with the blank control or alone treatment group,combination of sorafenib plus vitamin K2 could significantly inhibit the metastasis and tumor growth of human hepatocellular carcinoma cells in the liver or the lung.Maintenance treatment with sorafenib and vitamin K2 significantly improved the life quality and prolonged survival time of the recipient mice.Conclusion Combination of sorafenib plus vitamin K2 has synergistic therapeutic effects on human hepatocellular carcinoma,which can inhibit the liver or lung metastasis and prolong the survival time.
