SMAD4蛋白对胰腺癌细胞促凋亡作用的研究

摘　　要：SMAD4蛋白在嗜甲醇酵母中表达后 ,纯化鉴定。将纯化产物与两株胰腺癌细胞株共同孵育 ,用DNA序列梯图谱和TdT末端标记两种方法考察了此蛋白对体外培养的胰腺癌细胞生长的作用。结果显示SMAD4对胰腺癌细胞株SW1990有明显的促凋亡作用 。After SMAD4 was expressed in Pichia pastoris and characterized with immunologic assay and amino acid sequence in N terminal. The protein was co-incubated with pancreatic carcinoma cell lines, SW1990 and Capan Ⅱ,in order to test its effects of inducing apoptosis. With the DNA ladder and TdT assays, it showed that the protein was able to induce pancreatic carcinoma cell line SW1990 apoptosis, while it had no effect on pancreatic carcinoma Capan Ⅱ cell line.
