骨髓成骨细胞在合成碳酸磷灰石表面的分化增殖研究

摘  要:目的:研究骨髓基质细胞转化的成骨细胞在磷酸盐溶液浸泡法合成的碳酸磷灰石(fabricated carbonate apatite,CAp)上的分化增殖。方法:通过Ca(OH)2的碳酸化生成的CaCO3浸泡在60℃、1 mol/L磷酸氢二钠盐溶液中14 d后,合成具有优良机械性能的低结晶性B型CAp团块。应用骨髓成骨细胞作为实验工具,观察体外细胞在CAp及作为对照组的烧结羟基磷灰石(sintered hydroxide apatite,HAP)、CaCO3、Ca(OH)2上的生长情况,定量分析碱性磷酸酶(alkalinephosphatase,ALP)及骨钙素(osteocalcin,OC)在成骨过程中不同时期的表达,并通过扫描电子显微镜(SEM)对细胞形态进行观察。结果:在早期培养7 h后,CAp上细胞贴壁率高于对照组;随后细胞不断增殖,在7 d后生长细胞数目呈现为:烧结HAP>CAp>CaCO3,而成骨细胞分化的标志——ALP活性与OC水平的表达也呈同样的趋势。SEM结果显示黏附在以上三种材料上的成骨细胞形态无明显差异。结论:通过磷酸盐溶液浸泡法合成的CAp能够促进体外培养成骨细胞黏附与增殖,有利于细胞向成骨分化,促进骨形成,有望成为理想的人工骨材料。Objective: To investigate the osteoblast differentiation of rat bone marrow cells on the fabricated carbonate apatite (CAp) treated with phosphate solutions. Methods: Calcium carbonate (CaCO3 ) blocks were prepared by carbonation of calcium hydroxide [ Ca (OH)2 ] blocks were treated in disodium hydrogen phosphate (Na2HPO4 ) solutions and fabicated a low-crystalline B- type CAp blocks with adequate strength. Cell behavior, alkaline phosphatase (ALP) activity, osteocalcin (OC) and cell morphology on CAp blocks were compared with those on sintered hydroxide apatite (HAP), prepared CAp, CaCO3 and Ca (OH)2 blocks as controls in vitro. Results: Osteoblast attached better on CAp than controls, and proliferated graduately with time, the cell number on the specimens was in the order of sintered HAP 〉 CAp 〉 CaCO3 after 7 d. As early and later osteobalst differentiation marks, ALP activity and OC result showed the same trend as proliferation rate. No significant difference could be seen by SEM observation. Conclusions: The fabricated CAp blocks has good biocompatibility with osteoblast, can enhance the osteoblast differentiation well and be regarded as a promising bone substitutes.
